Regulation of c-Maf and {alpha}A-crystallin in Ocular Lens by FGF Signaling Developmental Biology

Fibroblast growth factor (FGF) signaling regulates a multitude of cellular processes, including cell proliferation, survival, migration, and differentiation. In the vertebrate lens, FGF signaling regulates fiber cell differentiation characterized by high expression of crystallin proteins. However, a direct link between FGF signaling and crystallin gene transcriptional machinery remains to be established. Previously, we have shown […]

Structural and mechanistic insights into the regulation of the fundamental Rho-regulator RhoGDI{alpha} by lysine acetylation Protein Structure and Folding

Rho-proteins are small GTP/GDP-binding proteins primarily involved in cytoskeleton regulation. Their GTP/GDP-cycle is often tightly connected to a membrane/cytosol-cycle regulated by the Rho guanine nucleotide dissociation inhibitor α (RhoGDIα). RhoGDIα has been regarded as a housekeeping regulator essential to control homeostasis of Rho-proteins. Recent proteomic screens showed that RhoGDIα is extensively lysine acetylated. Here, we […]

Distinct Developmental Functions of Prostasin (CAP1/PRSS8) Zymogen and Activated Prostasin Enzymology

The membrane-anchored serine prostasin (CAP1/PRSS8) is essential for barrier acquisition of the interfollicular epidermis and for normal hair follicle development. Consequently, prostasin null mice die shortly after birth. Prostasin is found in two forms in the epidermis: a one-chain zymogen and a two-chain proteolytically active form, generated by matriptase-dependent activation site cleavage. Here we used […]

The E3-ubiquitin ligase adaptor protein Skp1 is glycosylated by an evolutionarily conserved pathway that regulates protist growth and development Microbiology

Toxoplasma gondii is a protist parasite of warm-blooded animals that causes disease by proliferating intracellularly in muscle and the central nervous system. Previous studies showed that a prolyl 4-hydroxylase related to animal HIFα prolyl hydroxylases is required for optimal parasite proliferation, especially at low O2. We also observed that Pro154 of Skp1, a subunit of […]

Molecular Pathways: Mechanism of Action for Talimogene Laherparepvec, a New Oncolytic Virus Immunotherapy

Oncolytic viruses are native or engineered viruses that preferentially replicate in and lyse cancer cells. Selective tumor cell replication is thought to depend on infection of neoplastic cells, which harbor low levels of protein kinase R (PKR) and dysfunctional type 1 interferon signaling elements. These changes allow more efficient viral replication, and with selected deletion […]
