Specificity of the Human Frequency Following Response for Carrier and Modulation Frequency Assessed Using Adaptation

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 1 λεπτό
Specificity of the Human Frequency Following Response for Carrier and Modulation Frequency Assessed Using Adaptation: AbstractThe frequency following response (FFR) is a scalp-recorded measure of phase-locked brainstem activity to stimulus-related periodicities. Three experiments investigated the specificity of the FFR for carrier and modulation frequency using adaptation. FFR waveforms evoked by alternating-polarity stimuli were averaged for each polarity and added, to enhance envelope, or subtracted, to enhance temporal fine structure information. The first experiment investigate...περισσότερα »

Repeated Moderate Noise Exposure

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 2 δευτερόλεπτα
Repeated Moderate Noise Exposure in the Rat—an Early Adulthood Noise Exposure Model: AbstractIn this study, we investigated the effects of varying intensity levels of repeated moderate noise exposures on hearing. The aim was to define an appropriate intensity level that could be repeated several times without giving rise to a permanent hearing loss, and thus establish a model for early adulthood moderate noise exposure in rats. Female Sprague-Dawley rats were exposed to broadband noise for 90 min, with a 50 % duty cycle at levels of 101, 104, 107, or 110 dB sound pressure level (SP...περισσότερα »

Nonlinear Vibration Response Measured at Umbo and Stapes in the Rabbit Middle ear

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 2 δευτερόλεπτα
Nonlinear Vibration Response Measured at Umbo and Stapes in the Rabbit Middle ear: AbstractUsing laser vibrometry and a stimulation and signal analysis method based on multisines, we have measured the response and the nonlinearities in the vibration of the rabbit middle ear at the level of the umbo and the stapes. With our method, we were able to detect and quantify nonlinearities starting at sound pressure levels of 93-dB SPL. The current results show that no significant additional nonlinearity is generated as the vibration signal is passed through the middle ear chain. Nonlineari... περισσότερα »

Temporal Resolution of the Normal Ear in Listeners with Unilateral Hearing Impairment

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 3 δευτερόλεπτα
Temporal Resolution of the Normal Ear in Listeners with Unilateral Hearing Impairment: AbstractUnilateral hearing loss (UHL) leads to an imbalanced input to the brain and results in cortical reorganization. In listeners with unilateral impairments, while the perceptual deficits associated with the impaired ear are well documented, less is known regarding the auditory processing in the unimpaired, clinically normal ear. It is commonly accepted that perceptual consequences are unlikely to occur in the normal ear for listeners with UHL. This study investigated whether the temporal res... περισσότερα »

Finite-Element Modelling of the Response of the Gerbil Middle Ear to Sound

Finite-Element Modelling of the Response of the Gerbil Middle Ear to Sound: AbstractWe present a finite-element model of the gerbil middle ear that, using a set of baseline parameters based primarily on a priori estimates from the literature, generates responses that are comparable with responses we measured in vivo using multi-point vibrometry and with those measured by other groups. We investigated the similarity of numerous features (umbo, pars-flaccida and pars-tensa displacement magnitudes, the resonance frequency and break-up frequency, etc.) in the experimental responses with...περισσότερα »

Concurrent Acoustic Activation of the Medial Olivocochlear System Modifies the After-Effects of Intense Low-Frequency Sound on the Human Inner Ear

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 4 δευτερόλεπτα
Concurrent Acoustic Activation of the Medial Olivocochlear System Modifies the After-Effects of Intense Low-Frequency Sound on the Human Inner Ear: Abstract>Human hearing is rather insensitive for very low frequencies (i.e. below 100 Hz). Despite this insensitivity, low-frequency sound can cause oscillating changes of cochlear gain in inner ear regions processing even much higher frequencies. These alterations outlast the duration of the low-frequency stimulation by several minutes, for which the term ‘bounce phenomenon’ has been coined. Previously, we have shown that the bounce can...περισσότερα »

Channel Interaction and Current Level Affect Across-Electrode Integration of Interaural Time Differences in Bilateral Cochlear-Implant Listeners

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 4 δευτερόλεπτα
Channel Interaction and Current Level Affect Across-Electrode Integration of Interaural Time Differences in Bilateral Cochlear-Implant Listeners: AbstractSensitivity to interaural time differences (ITDs) is important for sound localization. Normal-hearing listeners benefit from across-frequency processing, as seen with improved ITD thresholds when consistent ITD cues are presented over a range of frequency channels compared with when ITD information is only presented in a single frequency channel. This study aimed to clarify whether cochlear-implant (CI) listeners can make use of s... περισσότερα »

Effects of Spectral Degradation on Attentional Modulation of Cortical Auditory Responses to Continuous Speech

Effects of Spectral Degradation on Attentional Modulation of Cortical Auditory Responses to Continuous Speech: AbstractThis study investigates the effect of spectral degradation on cortical speech encoding in complex auditory scenes. Young normal-hearing listeners were simultaneously presented with two speech streams and were instructed to attend to only one of them. The speech mixtures were subjected to noise-channel vocoding to preserve the temporal envelope and degrade the spectral information of speech. Each subject was tested with five spectral resolution conditions (unprocess...περισσότερα »

Loss of Afferent Vestibular Input Produces Central Adaptation and Increased Gain of Vestibular Prosthetic Stimulation

Loss of Afferent Vestibular Input Produces Central Adaptation and Increased Gain of Vestibular Prosthetic Stimulation: ABSTRACTImplanted vestibular neurostimulators are effective in driving slow phase eye movements in monkeys and humans. Furthermore, increases in slow phase velocity and electrically evoked compound action potential (vECAP) amplitudes occur with increasing current amplitude of electrical stimulation. In intact monkeys, protracted intermittent stimulation continues to produce robust behavioral responses and preserved vECAPs. In lesioned monkeys, shorter duration stud... περισσότερα »

Hair Cell Loss, Spiral Ganglion Degeneration, and Progressive Sensorineural Hearing Loss in Mice with Targeted Deletion of Slc44a2/Ctl2: AbstractSLC44A2 (solute carrier 44a2), also known as CTL2 (choline transporter-like protein 2), is expressed in many supporting cell types in the cochlea and is implicated in hair cell survival and antibody-induced hearing loss. In mice with the mixed C57BL/6-129 background, homozygous deletion of *Slc44a2* exons 3–10 (*Slc44a2* Δ/Δ) resulted in high-frequency hearing loss and hair cell death. Tuning to Binaural Cues in Human Auditory Cortex

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 4 δευτερόλεπτα
Tuning to Binaural Cues in Human Auditory Cortex: AbstractInteraural level and time differences (ILD and ITD), the primary binaural cues for sound localization in azimuth, are known to modulate the tuned responses of neurons in mammalian auditory cortex (AC). The majority of these neurons respond best to cue values that favor the contralateral ear, such that contralateral bias is evident in the overall population response and thereby expected in population-level functional imaging data. Human neuroimaging studies, however, have not consistently found contralaterally biased binaural... περισσότερα »

Temporal Considerations for Stimulating Spiral Ganglion Neurons with Cochlear Implants

Temporal Considerations for Stimulating Spiral Ganglion Neurons with Cochlear Implants: AbstractA wealth of knowledge about different types of neural responses to electrical stimulation has been developed over the past 100 years. However, the exact forms of neural response properties can vary across different types of neurons. In this review, we survey four stimulus-response phenomena that in recent years are thought to be relevant for cochlear implant stimulation of spiral ganglion neurons (SGNs): refractoriness, facilitation, accommodation, and spike rate adaptation. Diagnosis of Cochlear Neuropathy with Envelope Following Responses

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 4 δευτερόλεπτα
Towards a Diagnosis of Cochlear Neuropathy with Envelope Following Responses Abstract Listeners with normal audiometric thresholds can still have suprathreshold deficits, for example, in the ability to discriminate sounds in complex acoustic scenes. One likely source of these deficits is cochlear neuropathy, a loss of auditory nerve (AN) fibers without hair cell damage, which can occur due to both aging and moderate acoustic overexposure. Since neuropathy can affect up to 50 % of AN fibers, its impact on suprathreshold hearing is likely profound, but progress is hindered by lack of a ... περισσότερα »

Stimulus Frequency Otoacoustic Emission Delays and Generating Mechanisms

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 8 δευτερόλεπτα
Stimulus Frequency Otoacoustic Emission Delays and Generating Mechanisms in Guinea Pigs, Chinchillas, and Simulations Abstract According to coherent reflection theory (CRT), stimulus frequency otoacoustic emissions (SFOAEs) arise from cochlear irregularities coherently reflecting energy from basilar membrane motion within the traveling-wave peak. This reflected energy arrives in the ear canal predominantly with a single delay at each frequency. However, data from humans and animals indicate that (1) SFOAEs can have multiple delay components, (2) low-frequency SFOAE delays are too sh... περισσότερα »

Selective Inner Hair Cell Dysfunction Impairs Hearing-in-Noise in the Absence of Outer Hair Cell Loss

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 9 δευτερόλεπτα
Selective Inner Hair Cell Dysfunction in Chinchillas Impairs Hearing-in-Noise in the Absence of Outer Hair Cell Loss Abstract Poorer hearing in the presence of background noise is a significant problem for the hearing impaired. Ototoxic drugs, ageing, and noise exposure can damage the sensory hair cells of the inner ear that are essential for normal hearing sensitivity. The relationship between outer hair cell (OHC) loss and progressively poorer hearing sensitivity in quiet or in competing background noise is supported by a number of human and animal studies. In contrast, the effect ... περισσότερα »

Increasing development of antimicrobial resistance is driving a resurgence in interest in phage therapy: the use of bacteriophages to treat bacterial infections. As the lytic action of bacteriophages is unaffected by the antibiotic resistance status of their bacterial target, it is thought that phage therapy may have considerable potential in the treatment of a wide range of topical and localized infections. As yet this interest has not extended to intravenous (IV) use, which is surprising given that the historical record shows that phages are likely to be safe and effective when ... περισσότερα »

Identification of DNA methylation-independent epigenetic events underlying clear cell renal cell carcinoma

Alterations in chromatin accessibility independent of DNA methylation can affect cancer-related gene expression, but are often overlooked in conventional epigenomic profiling approaches. In this study, we describe a cost-effective and computationally simple assay called AcceSssIble to simultaneously interrogate DNA methylation and chromatin accessibility alterations in primary human clear cell renal cell carcinomas (ccRCC). Our study […]

AKT1 inhibits epithelial-to-mesenchymal transition in breast cancer through phosphorylation-dependent Twist1 degradation

Epithelial-to-mesenchymal transition (EMT) is an essential physiological process that promotes cancer cell migration, invasion, and metastasis. Several lines of evidence from both cellular and genetic studies suggest that AKT1/PKBα, but not AKT2 or AKT3, serves as a negative regulator of EMT and breast cancer metastasis. However, the underlying mechanism by which AKT1 suppresses EMT remains […]

Preclinical anticancer efficacy of BET bromodomain inhibitors is determined by the apoptotic response

Small molecule inhibitors of the bromodomain and extraterminal (BET) family of proteins are being tested in clinical trials for a variety of cancers, but patient selection strategies remain limited. This challenge is partly attributed to the heterogeneous responses elicited by BET inhibition (BETi), including cellular differentiation, senescence, and death. In this study, we performed phenotypic […]

Caution Against Overinvestigation of Small Thyroid Nodules: *In Reply* We would like to thank the radiologists who wrote a letter in response to our recently published article. Their primary concern was that our finding of smaller thyroid nodules (TNs) having a relatively increased likelihood of malignancy regardless of Bethesda classification could be misconstrued by readers and, in so doing, perpetuate the overinvestigation of TN. This concern is indeed valid, as this is a true phenomenon within our field.

Abstract Objectives Hepatic steatosis is common but less is known of the heterogeneity of hepatic fat distribution and its clinical significance. Our objective was to measure the regional variabilities within the liver of standardised uptake values (SUV) as potential markers of hepatic fat distribution heterogeneity. Methods Twenty-four patients having routine, clinically indicated PET/CT with 18F-FDG and a wide range of fatty liver severity were selected. Maximum SUV (SUVmax), average SUV (SUVave), both calculated using lean body mass, and CT density were measured in 12 × 2-cm d... περισσότερα »

Cyclin D1 and DNA Damage Sensing

Therapy resistance and poor outcome in prostate cancer is associated with increased expression of cyclin D1. Androgens promote DNA double-strand break repair to reduce DNA damage, and cyclin D1 was also shown to enhance DNA damage repair (DDR). In this study, we investigated the significance of cyclin D1 in androgen-induced DDR using established prostate cancer […]

IL21 Signaling in Anti-ErbB2 Therapy

The HER2/ErbB2 monoclonal antibody (mAb) trastuzumab is combined with chemotherapy as a standard-of-care for newly diagnosed HER2+ breast cancer patients, but some patients treated with this combination therapy experience early relapse. Our analysis of data from a clinical trial evaluating the efficacy of chemotherapy plus/minus trastuzumab suggested that the magnitude of trastuzumab benefit on distant […]

KIR and HLA in Skin Cancer

Natural killer (NK)–cell phenotype is partially mediated through binding of killer-cell immunoglobulin-like receptors (KIR) with HLA class I ligands. The KIR gene family is highly polymorphic and not well captured by standard genome-wide association study approaches. Here, we tested the hypothesis that variations in KIR gene content combined with HLA class I ligand status is […]

PRC2, Th1 Chemokines, and Tumor Immunity

Infiltration of tumors with effector T cells is positively associated with therapeutic efficacy and patient survival. However, the mechanisms underlying effector T-cell trafficking to the tumor microenvironment remain poorly understood in patients with colon cancer. The polycomb repressive complex 2 (PRC2) is involved in cancer progression, but the regulation of tumor immunity by epigenetic mechanisms […]

PD-1-Mediated Paralysis of DCs

The PD-1:PD-L1 immune signaling axis mediates suppression of T-cell–dependent tumor immunity. PD-1 expression was recently found to be upregulated on tumor-infiltrating murine (CD11c+CD11b+CD8−CD209a+) and human (CD1c+CD19−) myeloid dendritic cells (TIDC), an innate immune cell type also implicated in immune escape. However, there is little knowledge concerning how PD-1 regulates innate immune cells. In this study, […]

MYC and Pten Cooperate to Induce Prostate Cancer

Genetic instability, a hallmark feature of human cancers including prostatic adenocarcinomas, is considered a driver of metastasis. Somatic copy number alterations (CNA) are found in most aggressive primary human prostate cancers, and the overall number of such changes is increased in metastases. Chromosome 10q23 deletions, encompassing PTEN, and amplification of 8q24, harboring MYC, are frequently […]

Guanylin Suppression by Calorie-Induced ER Stress in Obesity

Obesity is a well-known risk factor for colorectal cancer but precisely how it influences risks of malignancy remains unclear. During colon cancer development in humans or animals, attenuation of the colonic cell surface receptor guanylyl cyclase C (GUCY2C) that occurs due to loss of its paracrine hormone ligand guanylin contributes universally to malignant progression. In […]

HBXIP and LSD1 Scaffolded by Hotair Activate c-Myc

c-Myc is regarded as a transcription factor, but the basis for its function remains unclear. Here, we define a long noncoding RNA (lncRNA)/protein complex that mediates the transcriptional activation by c-Myc in breast cancer cells. Among 388 c-Myc target genes in human MCF-7 breast cancer cells, we found that their promoters could be occupied by […]

BRAF/PI3K{beta}/IGF1R Inhibitor Synergy in PTENLOF/BRAFMUT Melanoma

The introduction of MAPK pathway inhibitors paved the road for significant advancements in the treatment of BRAF-mutant (BRAFMUT) melanoma. However, even BRAF/MEK inhibitor combination therapy has failed to offer a curative treatment option, most likely because these pathways constitute a codependent signaling network. Concomitant PTEN loss of function (PTENLOF) occurs in approximately 40% of BRAFMUT […]

Melanoma Phenotype-Dependent Mast Cell Recruitment

Human melanomas exhibit considerable genetic, pathologic, and microenvironmental heterogeneity. Anatomical variants that affect both the anterior and posterior digastric muscles are well described, and include duplications and variable origin or sites of insertion. Although agenesis or atrophy of the anterior digastric occurs in up to 40% of patients with hemifacial microsomia, to our knowledge, it has been reported only rarely in those without this syndrome. To minimise observer bias and to establish the incidence of anomalies and atrophy of this muscle, a prospective 6-year study was conducted by an experienced consultant radiologist in all patients referred for ultrasound ... περισσότερα »
