Breast, ovarian, and prostate cancers are hormone-related and may have a shared genetic basis, but this has not been investigated systematically by genome-wide association (GWA) studies. Meta-analyses combining the largest GWA meta-analysis data sets for these cancers totaling 112,349 cases and 116,421 controls of European ancestry, all together and in pairs, identified at *P* < 10–8 seven new cross-cancer loci: three associated with susceptibility to all three cancers (rs17041869/2q13/*BCL2L11*; rs7937840/11q12/*INCENP*; rs1469713/19p13/*GATAD2A*), two breast and ovarian cancer r... περισσότερα »

Atypical Rapidly Enlarging Orbital Schwannoma.
Atypical Rapidly Enlarging Orbital Schwannoma.: Orbital schwannomas are typically slow-growing benign tumors that can cause gradual loss of vision, proptosis, and limitation of ocular motility. The authors present an atypical case of a rapidly growing orbital apex schwannoma in a patient with preexisting vision loss secondary to presumed sarcoidal optic neuritis. Contrary to the slowly… Continue reading Atypical Rapidly Enlarging Orbital Schwannoma.

“Wild type” GIST: Clinicopathological features and clinical practice
Gastrointestinal stromal tumor (GIST) is a mesenchymal tumor of the gastrointestinal tract. Mutation of KIT and PDGFRA genes is implicated in the tumorigenesis. Approximately 10% of GISTs do not harbor mutation of these genes, and they are designated as «wild type» GIST. They are classified into succinate dehydrogenase (SDH)-deficient and non-SDH-deficient groups. SDH-deficient group includes… Continue reading “Wild type” GIST: Clinicopathological features and clinical practice
Simvastatin monotherapy as a potential option for statin-associated muscle symptoms: a case report
Summary What is known and objectives Choosing an alternative statin is recommended when managing statin-associated muscle symptoms (SAMS) and new hydrophilic statins are often suggested. We report on a case of statin-associated muscle damage that was successfully managed by simplifying the patient's combination therapy with simvastatin–ezetimibe to simvastatin alone. Case summary The patient experienced SAMS when he was successively treated with atorvastatin, rosuvastatin, Xuezhikang capsule and combined simvastatin/ezetimibe therapy. Gradual thrombocytopenia induced by long-term trastuzumab exposure
Summary What is known and objective Unexplained thrombocytopenia is a common clinical problem, and the possibility of drug-induced thrombocytopenia must be considered. Moreover, chemotherapy agents are known to typically cause thrombocytopenia by suppressing haematopoiesis, but they can also cause immune thrombocytopenia in which the mechanism involves antibody-mediated platelet destruction. Case description We report a case of severe thrombocytopenia related to trastuzumab administration in which the patient exhibits a slow drift downward in platelet counts with repeated cycles ... περισσότερα »

Anticancer drug discovery: Structures of KDM5 histone demethylase inhibitors
When doctors hurl toxic death at cancer cells, often a few will survive and come back. A family of enzymes called KDM5 histone demethylases is emerging as important for this resilience, and drugs that inhibit KDM5 enzymes could be active in treating several types of cancer. http://ift.tt/29OsyJa

Berberine Inhibits Intestinal Polyps Growth in Apc (min/+) Mice via Regulation of Macrophage Polarization
Antitumor effect of berberine has been reported in a wide spectrum of cancer, however, the mechanisms of which are not fully understood. The aim of this study was to investigate the hypothesis that berberine suppresses tumorigenesis in the familial adenomatous polyposis (FAP) by regulating the macrophage polarization in Apc (min/+) mouse model. Berberine was given to Apc (min/+) mice for 12 weeks. Primary macrophages were isolated; after berberine treatment, the change in signaling cascade was determined. The total number and size of polyps were reduced remarkably in berberine grou... περισσότερα »

Circulating MicroRNAs as Potential Molecular Biomarkers in Pathophysiological Evolution of Pregnancy
MicroRNAs represent nonprotein coding small RNA molecules that are very stable to degradation and responsible for gene silencing in most eukaryotic cells. Increased evidence has been accumulating over the years about their potential value as biomarkers for several diseases. MicroRNAs were predicted to be involved in nearly all biological processes from development to oncogenesis. In this review, we address the importance of circulating microRNAs in different conditions associated with pregnancy starting with the implantation period to preeclampsia and we shortly describe the corre... περισσότερα »

Gastrointestinal stromal tumor (GIST) is a mesenchymal tumor of the gastrointestinal tract. Mutation of *KIT* and *PDGFRA* genes is implicated in the tumorigenesis. Approximately 10% of GISTs do not harbor mutation of these genes, and they are designated as "wild type" GIST. They are classified into succinate dehydrogenase (SDH)-deficient and non-SDH-deficient groups. SDH-deficient group includes Carney triad and Carney Stratakis syndrome. The patients are young women. Tumors occur in the antrum of the stomach, and tumor cells are epithelioid. Lymph node metastasis is frequent. T... περισσότερα »

[image: cover.gif?v=1&s=21833297c1367a913fe9a230] Aims Little is known about the impact of Inappropriate Prescribing (IP) in community-dwelling adults, aged 80 and older. The prevalence at baseline (November 2008 – September 2009) and impact of IP (misuse, and underuse) after 18 months on mortality, and hospitalisation in a cohort of community-dwelling adults, aged 80 and older (n = 503) was studied. Hypoglycaemia when Adding Sulphonylurea to Metformin: A Systematic Review and Network Meta-analysis
Aims/hypothesis The risk of hypoglycaemia may differ among sulphonylureas (SUs), but evidence from head-to-head comparisons is sparse. Performing a network meta-analysis to use indirect evidence from randomised controlled trials (RCTs), we compared the relative risk of hypoglycaemia with newer generation SUs when added to metformin. Methods A systematic review identified RCTs lasting 12-52 weeks and evaluating SUs added to inadequate metformin monotherapy (≥1000 mg/day) in type 2 diabetes. Adding RCTs investigating the active comparators from the identified SU-trials, we establi... περισσότερα »
