7-Rhamnosylated Flavonols Modulate Homeostasis of the Plant Hormone Auxin and Affect Plant Development Developmental Biology

Flavonols are a group of secondary metabolites that affect diverse cellular processes. They are considered putative negative regulators of the transport of the phytohormone auxin, by which they influence auxin distribution and concomitantly take part in the control of plant organ development. Flavonols are accumulating in a large number of glycosidic forms. Whether these have […]

Filamin A Is Involved in HIV-1 Vpu Mediated Evasion of Host Restriction by Modulating Tetherin Expression Cell Biology

Tetherin, also known as bone marrow stromal antigen 2 (BST-2), inhibits the release of a wide range of enveloped viruses including HIV-1 by directly tethering nascent virions to the surface of infected cells. HIV-1 accessary protein Vpu counteracts tetherin restriction via sequestration, downregulation, and/or displacement mechanisms to remove tetherin from the sites of virus budding. […]

Evolutionary Ancestry of Eukaryotic Protein Kinases and Choline Kinases Computational Biology

The reversible phosphorylation of proteins catalyzed by protein kinases in eukaryotes supports an important role for eukaryotic protein kinases (ePKs) in the emergence of nucleated cells in the third superkingdom of life. Choline kinases (ChKs) could also be critical in the early evolution of eukaryotes, because of their function in the biosynthesis of phosphatidylcholine, which […]

Targeted Next-Generation Sequencing Analysis of a Pendred Syndrome-Associated Thyroid Carcinoma

Rhizobiale-like phosphatase 2 from Arabidopsis thaliana is a novel phospho-tyrosine specific PPP-family protein phosphatase Signal Transduction

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 1 ώρα
Cellular signaling through protein tyrosine phosphorylation is well established in mammalian cells. Although lacking the classic tyrosine kinases present in humans, plants have a tyrosine phosphoproteome that rivals human cells. Here we report a novel plant tyrosine phosphatase from Arabidopsis thaliana (AtRLPH2) that surprisingly, has the sequence hallmarks of a phospho-serine/threonine phosphatase belonging to the… Continue reading Rhizobiale-like phosphatase 2 from Arabidopsis thaliana is a novel phospho-tyrosine specific PPP-family protein phosphatase Signal Transduction

Conserved amphipathic helices mediate lipid droplet targeting of perilipins 1-3. Cell Biology

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 1 ώρα
Perilipins (PLINs) play a key role in energy storage by orchestrating the activity of lipases on the surface of lipid droplets (LDs). Failure of this activity results in severe metabolic disease in humans. Unlike all other lipid droplet associated proteins, PLINs localise almost exclusively to the phospholipid monolayer surrounding the droplet. To understand how they… Continue reading Conserved amphipathic helices mediate lipid droplet targeting of perilipins 1-3. Cell Biology

Identification of glycosylation sites essential for surface expression of the CaV{alpha}2{delta} 1 subunit and modulation of the cardiac CaV1.2 channel activity Protein Structure and Folding

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 1 ώρα
Alterations in the L-type current density is one aspect of the electrical remodeling observed in patients suffering from cardiac arrhythmias. Changes in channel function could result from variations in the protein biogenesis, stability, post-translational modification, and/or trafficking in any of the regulatory subunits forming cardiac L-type Ca2+ channel complexes. CaVα2δ1 is potentially the most heavily… Continue reading Identification of glycosylation sites essential for surface expression of the CaV{alpha}2{delta} 1 subunit and modulation of the cardiac CaV1.2 channel activity ... περισσότερα »

NusG is a Sequence-specific RNA Polymerase Pause Factor that Binds to the Non-template DNA Within the Paused Transcription Bubble Microbiology

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 1 ώρα
NusG, referred to as Spt5 in archaeal and eukaryotic organisms, is the only transcription factor conserved in all three domains of life. This general transcription elongation factor binds to RNA polymerase (RNAP) soon after transcription initiation and dissociation of the RNA polymerase σ factor. Escherichia coli NusG increases transcription processivity by suppressing RNAP pausing, whereas… Continue reading NusG is a Sequence-specific RNA Polymerase Pause Factor that Binds to the Non-template DNA Within the Paused Transcription Bubble Microbiology

Heterogeneity of Collagen VI Microfibrils: Structural Analysis of Non-collagenous Regions Glycobiology and Extracellular Matrices

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 1 ώρα
Collagen VI, a collagen with uncharacteristically large N- and C-terminal non-collagenous regions, forms a distinct microfibrillar network in most connective tissues. It was long considered to consist of three genetically distinct α chains (α1, α2 and α3). Intracellularly heterotrimeric molecules associate to form dimers and tetramers, which are then secreted and assembled to microfibrils. The… Continue reading Heterogeneity of Collagen VI Microfibrils: Structural Analysis of Non-collagenous Regions Glycobiology and Extracellular Matrices

How generalizable are the results from trials of Direct Antiviral Agents to people coinfected with HIV/Hepatitis C virus in the real world?

Alexandros G. SfakianakisστοΛοίμωξηΠριν από 1 ώρα
Trial results are used to support licensure, inform cost-effectiveness analyses and guide clinical decision-making. We found the majority of co-infected patients were not included in clinical trials of DAAs, raising concerns about the generalizability of these trial results. from #Infections via alexandrossfakianakis on Inoreader http://ift.tt/1S7tHMe via IFTTT

Complex mode of inheritance in holoprosencephaly revealed by whole exome sequencing

Alexandros G. SfakianakisστοEvolution of a geneΠριν από 1 ώρα
Abstract Holoprosencephaly (HPE) is the most common congenital cerebral malformation, characterized by impaired forebrain cleavage and midline facial anomalies. Heterozygous mutations in 14 genes have been associated with HPE, and are often inherited from an unaffected parent underlying complex genetic bases. It is now emerging that HPE may result from a combination of multiple genetic events, rather than from a single heterozygous mutation. To explore this hypothesis, we undertook whole exome sequencing (WES) and targeted high-throughput sequencing approaches to identify mutation...περισσότερα »

The c.65-2A>G splice site mutation is associated with a mild phenotype in Danon disease due to the transcription of normal LAMP2 mRNA

Alexandros G. SfakianakisστοEvolution of a geneΠριν από 1 ώρα
Abstract Danon disease (DD) is a rare X-linked multisystem disorder caused by mutations of the LAMP2 gene and characterized by intellectual disability, skeletal myopathy and cardiomyopathy. The survival time is severely reduced. Contrasting with the usual disease course we report on a family with an exceptionally mild phenotype of DD despite having two potentially damaging LAMP2 mutations. Using RNA-Seq analysis we showed that a c.65-2A>G splice site mutation results in the tissue-specific production of four different transcripts including the full-length mRNA in muscle tissue but ...περισσότερα »
