Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 26 δευτερόλεπτα
Disparate Response to Methotrexate in Stem Versus Non-Stem Cells: AbstractMethotrexate (MTX) is a commonly used chemotherapeutic agent that kills cancer cells by binding dihydrofolate reductase (DHFR) as a competitive inhibitor. Due to its non-selectivity, MTX also impairs normal (non-cancerous) cell function and causes long-term damage to healthy tissue. These consequences have been investigated extensively in bone-derived cells due to their sensitivity to the drug. While DHFR likely plays a role in normal cell response to MTX, research in this area is limited. Moreover, how MTX s...περισσότερα »

Replacement of Lost Positive Stem Cells through Plasticity of Their Enterocyte-Lineage Daughters

Replacement of Lost Positive Stem Cells through Plasticity of Their Enterocyte-Lineage Daughters: [image: article_notepad.gif] In this article, Tetteh et al. show that enterocyte-lineage progenitors can become stem cells during intestinal regeneration. Additionally, these cells generate Paneth-like cells and turn on genes that promote recovery from injury. In sum, “stemness” in intestinal crypts is not “hard-wired;” many progenitors can regain stemness upon loss of the actual stem cells.

Alexandros G. SfakianakisστοΟγκολογίαΠριν από 36 δευτερόλεπτα
Abstract The extracellular signal-regulated kinase 1/2 (ERK1/2) signaling pathway is activated by several growth factors and mitogens, and upregulation has been noted in many human cancers, including examples in the lung. In this study, to study the association of ERK1/2 activation with mutation of *Kras* encoding an upstream activator of ERK1/2 in lung premalignant lesions, we immunohistochemically examined expression of phosphorylated forms of ERK1/2 (pERK1/2) and MAP/ERK kinase 1/2 (pMEK1/2) proteins and correlation between ERK activation and mutation of *Kras* encoding an upstr...περισσότερα »

MicroRNA-21 induces 5-fluorouracil resistance in human pancreatic cancer cells by regulating PTEN and PDCD4

Alexandros G. SfakianakisστοΟγκολογίαΠριν από 36 δευτερόλεπτα
Abstract Pancreatic cancer patients are often resistant to chemotherapy treatment, which results in poor prognosis. The objective of this study was to delineate the mechanism by which miR-21 induces drug resistance to 5-fluorouracil (5-FU) in human pancreatic cancer cells (PATU8988 and PANC-1). We report that PATU8988 cells resistant to 5-FU express high levels of miR-21 in comparison to sensitive primary PATU8988 cells. Suppression of miR-21 expression in 5-Fu-resistant PATU8988 cells can alleviate its 5-FU resistance. Meanwhile, lentiviral vector-mediated overexpression of miR-2...περισσότερα »

Is tasting innate? Authors: Spielman AI, Brand J Abstract Unique features of the sense of taste have engendered two major hypotheses on how the brain recognizes and classifies taste stimuli. These two hypotheses are both based on the same data set: the ability to recognize a large number of stimuli, yet to perceptually classify each within 5 primary qualities. A point of contention for years has been the manner in which our brain accomplishes this task. One idea considers that perception involves the "reading" of a complex pattern or array of excited neurons. Because of the comple... περισσότερα »

Catalysts, Vol. 6, Pages 27: Highly Efficient Cationic Palladium Catalyzed Acetylation of Alcohols and Carbohydrate-Derived Polyols

Alexandros G. SfakianakisστοNews in OtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 55 δευτερόλεπτα
The development of a new facile method for the acetylation of alcohols and carbohydrate-derived polyols is described. This method relies on the nature of the cationic palladium catalyst, Pd(PhCN)2(OTf)2 which is generated in situ from Pd(PhCN)2Cl2 and AgOTf to catalyze the acetylation reaction. This new acetylation protocol is very rapid and proceeds under mild conditions with only 1 mol% of catalyst loading at room temperature. This new method has been applied to a variety of different alcohols with different levels of steric hindrance, as well as carbohydrate-derived polyols to p...περισσότερα »

Multikinase inhibitor regorafenib inhibits the growth and metastasis of colon cancer with abundant stroma

Abstract Interaction between tumor cells and stromal cells plays an important role in the growth and metastasis of colon cancer. We previously found that carcinoma-associated fibroblasts (CAFs) expressed platelet-derived growth factor receptor-beta (PDGFR-β) and that PDGFR targeted therapy by using imatinib or nilotinib inhibited stromal reaction. Bone marrow-derived mesenchymal stem cells (MSCs) migrate to tumor […]

The use of brief post-surgical low frequency electrical stimulation to enhance nerve regeneration in clinical practice

Alexandros G. SfakianakisστοNews in OtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 1 ώρα
Abstract Despite efforts to enhance peripheral nerve regeneration, there has been little progress in improving clinical outcomes. Recently, a method of brief post-surgical low frequency electrical stimulation of surgically repaired nerves has been developed. It was shown to accelerate axon outgrowth across the repair site and hastened target reinnervation. In this brief review, we describe the mechanistic insights and functional impacts of the post-surgical electrical stimulation that have been gained through animal studies. Brain derived neurotrophic factor, cyclic AMP and regen... περισσότερα »

Electrical synapses and the development of inhibitory circuits in the thalamus

Alexandros G. SfakianakisστοNews in OtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 1 ώρα
Abstract Neurons within the mature thalamic reticular nucleus (TRN) powerfully inhibit ventrobasal (VB) thalamic relay neurons via GABAergic synapses. TRN neurons are also coupled to one another by electrical synapses that depend strongly on the gap junction protein connexin36 (Cx36). Electrical synapses in the TRN precede the postnatal development of TRN-to-VB inhibition. We investigated how the deletion of Cx36 affects the maturation of TRN and VB neurons, electrical coupling, and GABAergic synapses by studying wild-type (WT) and Cx36 knockout (KO) mice. The incidence and streng...περισσότερα »

The repair Schwann cell and its function in regenerating nerves

Alexandros G. SfakianakisστοNews in OtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 1 ώρα
Abstract Nerve injury triggers the conversion of myelin and non-myelin (Remak) Schwann cells to a cell phenotype specialised to promote repair. Distal to damage, these repair Schwann cells provide the necessary signals and spatial cues for the survival of injured neurons, axonal regeneration and target reinnervation. The conversion to repair Schwann cells involves de-differentiation together with alternative differentiation, or activation, a combination that is typical of cell type conversions often referred to as (direct or lineage) reprogramming. Thus, injury-induced Schwann ce... περισσότερα »

Novel human three-domain antibody fragments against sTNFα as well as tmTNFα with high affinity generated by the combination of ribosome display and E.coli expression system

Abstract Human tumor necrosis factor α (hTNFα) has been proved to be a validated therapeutic target in a number of immune-mediated inflammatory diseases (IMIDs). Fully human monoclonal antibodies (mAbs) that can neutralize soluble hTNFα (sTNFα) as well as transmembrane hTNFα (tmTNFα) are more desirable hTNFα antagonists. Here we report that novel anti-hTNFα human low molecular […]
