HuR Mediates TRAIL Resistance

Alexandros G. SfakianakisστοSquamous cell carcinoma (SCC) - Cancer - MelanomaΠριν από 9 δευτερόλεπτα
Pancreatic ductal adenocarcinoma (PDA) is one of the most lethal cancers, in part, due to resistance to both conventional and targeted therapeutics. TRAIL directly induces apoptosis through engagement of cell surface Death Receptors (DR4 and DR5), and has been explored as a molecular target for cancer treatment. Clinical trials with recombinant TRAIL and DR-targeting agents, however, have failed to show overall positive outcomes. Herein, we identify a novel TRAIL resistance mechanism governed by Hu antigen R (HuR, ELAV1), a stress-response protein abundant and functional in PDA cel... περισσότερα »

DNA Repair Inhibition by Oxidative Stress

The relationship between sun exposure and nonmelanoma skin cancer risk is well established. Solar UV (wavelength 280–400 nm) is firmly implicated in skin cancer development. Nucleotide excision repair (NER) protects against cancer by removing potentially mutagenic DNA lesions induced by UVB (280–320 nm). How the 20-fold more abundant UVA (320–400 nm) component of solar UV radiation increases skin cancer risk is not understood. Here it is demonstrated that the contribution of UVA to the effect of UV radiation on cultured human cells is largely independent of its ability to damage DN... περισσότερα »

HNRNP E1 Posttranscriptionally Regulates CDC27 Expression

Alexandros G. SfakianakisστοSquamous cell carcinoma (SCC) - Cancer - MelanomaΠριν από 10 δευτερόλεπτα
CDC27 is a core component of the anaphase-promoting complex/cyclosome (APC/C), a multisubunit E3 ubiquitin ligase, whose oscillatory activity is responsible for the metaphase-to-anaphase transition and mitotic exit. Here, in normal murine mammary gland epithelial cells (NMuMG), CDC27 expression is controlled posttranscriptionally through the RNA binding protein poly(rC) binding protein 1 (PCBP1)/heterogeneous nuclear ribonucleoprotein E1 (HNRNP E1). shRNA-mediated knockdown of HNRNP E1 abrogates translational silencing of the *Cdc27* transcript, resulting in constitutive expressio... περισσότερα »

The Effect of Adipose Stem Cells on Breast Cancer

Alexandros G. SfakianakisστοSquamous cell carcinoma (SCC) - Cancer - MelanomaΠριν από 10 δευτερόλεπτα
Adipose-derived stem cells (ASC) have received considerable attention in oncology because of the known direct link between obesity and cancer as well as the use of ASCs in reconstructive surgery after tumor ablation. Previous studies have documented how cancer cells commandeer ASCs to support their survival by altering extracellular matrix composition and stiffness, migration, and metastasis. This study focused on delineating the effects of ASCs and adipocytes on the self-renewal of stem/progenitor cells and hierarchy of breast epithelial cells. The wingless (Wnt) signaling is suggested as a fundamental hierarchical pathway in regulation of proliferation and differentiation of cells. The Wnt ligands are small proteins of about 40 kDa essentially for regulation and initiation of the Wnt activity. They are secreted proteins requiring acylation for activity in the Wnt signaling cascade and for functional interactivity with transmembrane proteins. Dual lipidation is important for posttranslational activation of the overwhelming number of Wnt proteins and is probably involved in their spatial distribution. The intestinal mucosa... περισσότερα »

Background Oral lichen planus (OLP) and oral lichenoid lesions (OLL) are considered potentially malignant disorders with a cancer incidence of around 1% of cases, although this estimation is controversial. The aim of this study was to analyze the cancer incidence in a case series of patients with OLP and OLL and to explore clinicopathological aspects that may cause underestimation of the cancer incidence in these diseases. Methods A retrospective study was conducted of 102 patients diagnosed with OLP (*n* = 21, 20.58%) or OLL (*n* = 81) between January 2006 and January 2016. Pat... περισσότερα »

3-D View of Mutations May Identify Potential Targets for Cancer Drugs 07/14/2016 A new computational tool may help expand the known number of mutations in cancer cells that could be targeted with new or existing drugs. Researchers recently reported that the tool, called HotSpot3D, allowed them to model how genetic mutations change the ways proteins function and interact with each other to potentially drive cancer. Regulation of Inflammation by Lipid Mediators in Oral Diseases

στοOtoRhinoLaryngology - Head and Neck SurgeryΠριν από 59 δευτερόλεπτα
Abstract Lipid mediators (LM) of inflammation are a class of compounds derived from *ω*-3 and *ω*-6 fatty acids that play a wide role in modulating inflammatory responses. Some LM possess pro-inflammatory properties, while others possess pro-resolving characteristics, and the class switch from pro-inflammatory to pro-resolving is crucial for tissue homeostasis. In this article, we review the major classes of LM, focusing on their biosynthesis and signaling pathways, and their role in systemic and, especially, oral health and disease. We discuss the detection of these LM in various... περισσότερα »

From focal sepsis to periodontal medicine: A century of exploring the role of the oral microbiome in systemic disease

Abstract The oral microbiome is established within a few minutes after birth and forms stable multi-species communities that engage in a dynamic equilibrium with the host immune system. Dental caries, endodontic infections and periodontal diseases are bacterially driven diseases that are caused by dysbiotic microbiomes. Over a century ago, the focal infection theory implicated these infections in the etiology of several systemic diseases, ranging from arthritis to neurodegenerative diseases. However, a lack of concrete evidence, combined with the urgency with which clinicians embra... περισσότερα »

Left-right coordination from simple to extreme conditions during split-belt locomotion in the chronic spinal adult cat

Abstract Although left-right coordination is essential for locomotion, its control is poorly understood, particularly in adult mammals. To investigate the spinal control of left-right coordination, a spinal transection was performed in six adult cats that were then trained to recover hindlimb locomotion. Spinal cats performed tied-belt locomotion from 0.1 m s−1 to 1.0 m s−1 and split-belt locomotion with low to high (1:1.25-10) slow-fast speed ratios. With the left hindlimb stepping at 0.1 m s−1 and the right hindlimb stepping from 0.2 m s−1 to 1.0 m s−1, 1:1, 1:2, 1:3, 1:4 and 1:5... περισσότερα »

In vivo matching of postsynaptic excitability with spontaneous synaptic inputs during formation of the rat calyx of Held synapse

Abstract In the adult, principal neurons of the medial nucleus of the trapezoid body (MNTB) are typically contacted by a single, giant terminal called the calyx of Held, whereas during early development a principal neuron receives inputs from many axons. How these changes in innervation impact the postsynaptic activity has not yet been studied *in vivo*. We therefore recorded spontaneous inputs and intrinsic properties of principal neurons in anesthetized rat pups during the developmental period in which the calyx forms. A characteristic bursting pattern could already be observed ... περισσότερα »

Exogenous and endogenous angiotensin-ii decrease renal cortical oxygen tension in conscious rats by limiting renal blood flow

We hypothesised that both exogenous and endogenous angiotensin-II (AngII) can decrease the partial pressure of oxygen (pO2) in the renal cortex of unrestrained rats, which might in turn contribute to the progression of chronic kidney disease. Rats were instrumented with telemeters equipped with a carbon paste electrode for continuous measurement of renal cortical tissue pO2. The method reproducibly detected acute changes in cortical oxygenation induced by systemic hyperoxia and hypoxia. In conscious rats, renal cortical pO2 was dose-dependently reduced by intravenous AngII. Reduc... περισσότερα »

Congenital heart diseases and their association with the variant distribution features on susceptibility genes

Abstract Congenital heart disease (CHD), one cause of childhood morbidity and mortality, is mainly triggered by a combination of environmental and genetic factors. Several susceptible genes, such as *NKX2-5*, *GATA4* and *TBX5,* have been reported as closely related to heart and vessel development. CHD subtypes are classified by diverse clinical phenotypes such as atrial septal defects (ASD), ventricular septal defects (VSD), tetralogy of Fallot (TOF), and Holt-Oram syndrome (HOS). Here, we summarize the associations of the genetic variants in these three genes with CHD subtypes. ... περισσότερα »

Less evidence concerning the association between ambient temperature and mortality is available in developing countries/regions, especially inland areas of China, and few previous studies have compared the predictive ability of different temperature indictors (minimum, mean, and maximum temperature) on mortality. Indoleamine 2,3-dioxygenase (IDO) expression in dendritic cells (DCs) inhibits T-cell activation and promotes T-cell differentiation into regulatory T-cells. Moreover, IDO expression promotes resistance to immunotherapies targeting immune checkpoints such as the cytotoxic T lymphocyte antigen-4 (CTLA-4). Here, a patient with metastatic melanoma pretreated with ipilimumab, an anti-CTLA-4 blocking antibody, was vaccinated with IDO-silenced DCs cotransfected… Continue reading Diversification of Antitumour Immunity in a Patient with Metastatic Melanoma Treated with Ipilimumab and an IDO... περισσότερα »
