Update on Auditory Evoked Responses: Value of Chirp Stimuli in ABR/ASSR Measurement

Αλέξανδρος ΣφακιανάκηςστοMedicine by Alexandros G. SfakianakisΠριν από 1 λεπτό
When I first heard about chirps many years ago, none of the clinical ABR systems had chirps as stimuli. As time went on, some of the manufacturers began to introduce chirps. As with any other new development, they were heavily promoted. However, I was not going to start using a new type of stimulus when the existing stimuli - traditional tone bursts and clicks - worked just fine. I kept an open mind, but I looked for evidence. Today, there is plenty of evidence for the use of chirps. from #ORL-Sfakianakis via xlomafota13 on Inoreader http://ift.tt/297xR5r via IFTTT

Plasma taurine levels are not affected by vigabatrin in pediatric patients

Αλέξανδρος ΣφακιανάκηςστοMedicine by Alexandros G. SfakianakisΠριν από 1 λεπτό
Summary Vigabatrin is a highly effective antiseizure medication, but its use is limited due to concerns about retinal toxicity. One proposed mechanism for this toxicity is vigabatrin-mediated reduction of taurine. Herein we assess plasma taurine levels in a retrospective cohort of children with epilepsy, including a subset receiving vigabatrin. All children who underwent a plasma amino acid analysis as part of their clinical evaluation between 2006 and 2015 at Stanford Children's Health were included in the analysis. There were no significant differences in plasma taurine levels bet... περισσότερα »

MZ B cells migrate in a T-bet dependent manner and might contribute to the remission of collagen-induced arthritis by the secretion of IL-10

Alexandros G. SfakianakisστοAllergy Rhinology Immunology AsthmaΠριν από 3 δευτερόλεπτα
In mice, marginal zone (MZ) B cells are found principally in the MZ of the spleen and characterized as CD23-negative cells, primarily express polyreactive BCRs, high levels of complement receptor-2 and toll-like receptors (TLRs). Collagen-induced arthritis (CIA) is a commonly used animal model of human rheumatoid arthritis, considered as a Th1-mediated disease. Although the importance of MZ B cells in the initiation of CIA is well established, their role in remission is unexplored. Besides, playing a central role in Th1 cell development, T-bet has important functions in B cells. ... περισσότερα »

FoxP3 Expression in Macrophages, Cancer, and B Cells—Is It Real?

Alexandros G. SfakianakisστοAllergy Rhinology Immunology AsthmaΠριν από 3 δευτερόλεπτα
Abstract During the last decade, B regulatory cells are appreciated to have a central role in preventing autoimmunity and maintaining self-tolerance. They are characterized by expressing different phenotypic markers and the production of either IL-10 or TGF-β or both. The recent recognition of Fas ligand expressing B regulatory cells as "killer" cells established their role in maintaining viral persistence by preventing effective antiviral immune responses. The forkhead lineage-transcription factor (FoxP3) was considered for many years to be a highly specific intracellular regulato... περισσότερα »

Significance of Interleukin-6/STAT Pathway for the Gene Expression of REG Iα, a New Autoantigen in Sjögren’s Syndrome Patients, in Salivary Duct Epithelial Cells

Abstract The regenerating gene, *Reg*, was originally isolated from a rat regenerating islet complementary DNA (cDNA) library, and its human homologue was named *REG Iα*. Recently, we reported that *REG Iα* messenger RNA (mRNA), as well as its product, was overexpressed in ductal epithelial cells in the salivary glands of Sjögren's syndrome patients. Furthermore, autoantibodies against REG Iα were found in the sera of Sjögren's syndrome patients, and the patients who were positive for the anti-REG Iα antibody showed significantly lower saliva secretion than antibody-negative patie... περισσότερα »

Abstract Identification of allergen epitopes is a key component in proper understanding of the pathogenesis of type I allergies, for understanding cross-reactivity and for development of mimotope immunotherapeutics. Phage particles have garnered recognition in the field of molecular allergology due to their value not only in competitive immunoscreening of peptide libraries but also as immunogenic carriers of allergen mimotopes. They integrate epitope discovery technology and immunization functions into a single platform. This article provides an overview of allergen mimotopes iden... περισσότερα »

Disruption of de novo fatty acid synthesis via acetyl-CoA carboxylase 1 inhibition prevents acute graft-versus-host disease

Alexandros G. SfakianakisστοAllergy Rhinology Immunology AsthmaΠριν από 4 δευτερόλεπτα
[image: cover.gif?v=1&s=7ed1ee29e575b52989bb5c85] Abstract Upon antigen-specific or allogeneic activation, T cells sharply increase their metabolic activity to cope with augmented needs for proliferation and effector functions. Therefore, enzymes involved in energy metabolism constitute attractive targets to modulate the activity of pathogenic effector T cells in the setting of graft-versus-host-disease (GVHD). Here we show that T cells deficient for acetyl-CoA carboxylase 1 (TACC1) are dramatically less pathogenic than wild type T cells in a lethal C57BL/6 into BALB/c model of a... περισσότερα »

Post-arrest stalling rather than crawling favors CD8+ over CD4+ T-cell migration across the blood-brain barrier under flow in vitro

Although CD8+ T cells have been implied in the pathogenesis of multiple sclerosis (MS), the molecular mechanisms mediating CD8+ T-cell migration across the blood-brain barrier (BBB) into the central nervous system (CNS) are ill defined. Using in vitro live cell imaging, we directly compared the multi-step extravasation of activated CD4+ and CD8+ T cells across primary mouse brain microvascular endothelial cells (pMBMECs) as a model for the BBB under physiological flow. Significantly higher numbers of CD8+ than CD4+ T cells arrested on pMBMECs under non-inflammatory and inflammator... περισσότερα »

Selective depletion of CD11c+CD11b+ dendritic cells partially abrogates tolerogenic effects of intravenous MOG in murine EAE

Alexandros G. SfakianakisστοAllergy Rhinology Immunology AsthmaΠριν από 4 δευτερόλεπτα
Intravenous (i.v.) injection of a soluble myelin antigen can induce tolerance, which effectively ameliorates experimental autoimmune encephalomyelitis (EAE). We have previously shown that i.v. MOG induces tolerance in EAE and expands a subpopulation of tolerogenic CD11c+CD11b+ dendritic cells (DCs) with an immature phenotype having low expression of IA and co-stimulatory molecules CD40, CD86, and CD80. Here we further investigate the role of tolerogenic DCs in i.v. tolerance by injecting clodronate-loaded liposomes, which selectively deplete CD11c+CD11b+ and immature DCs, but not... περισσότερα »

SLAM-associated protein favours the development of iNKT2 over iNKT17 cells

Alexandros G. SfakianakisστοAllergy Rhinology Immunology AsthmaΠριν από 4 δευτερόλεπτα
[image: cover.gif?v=1&s=7ed1ee29e575b52989bb5c85] Invariant Natural Killer T (iNKT) cells differentiate in the thymus into three distinct lineages defined by their cytokine and transcription factor expression. SLAM-associated protein (SAP) is essential for early stages of iNKT cell development, but its role during terminal differentiation of iNKT1, iNKT2 or iNKT17 cells remains unclear. Taking advantage of SAP-deficient mice expressing a Vα14-Jα18 TCRα transgene, we found that SAP is critical not only for IL-4 production but also for the terminal differentiation of IL-4-producing ... περισσότερα »

Safeguarding Fertility With Whole Ovary Cryopreservation and Microvascular Transplantation: Higher Follicular Survival With Vitrification Than With Slow Freezing in a Ewe Model.

Alexandros G. SfakianakisστοAllergy Rhinology Immunology AsthmaΠριν από 4 δευτερόλεπτα
Background: In young women, ovarian cortex cryopreservation before gonadotoxic chemotherapy and its avascular grafting after cancer healing permitted fertility restoration. However, ischemia reduced the grafts' lifespan. Microvascular transplantation of cryopreserved whole ovary may allow immediate revascularization, ensuring better fertility preservation, but the best cryopreservation method is unknown. We aimed to compare slow freezing and vitrification of whole ovary for fertility preservation purposes, in an ewe model. Methods: Twelve ewes were allocated at random to slow freezi... περισσότερα »

Update on Auditory Evoked Responses: Value of Chirp Stimuli in ABR/ASSR Measurement

When I first heard about chirps many years ago, none of the clinical ABR systems had chirps as stimuli. As time went on, some of the manufacturers began to introduce chirps. As with any other new development, they were heavily promoted. Alexandros G. SfakianakisστοHead,Neck,Brain,Cervical Spine and Esophageal Cancers by Alexandros G. SfakianakisΠριν από 33 δευτερόλεπτα
