High immunosuppressive burden in cancer patients: a major hurdle for cancer immunotherapy

Alexandros G. SfakianakisστοOtoRhinoLaryngology - Alexandros G. SfakianakisΠριν από 8 δευτερόλεπτα
High immunosuppressive burden in cancer patients: a major hurdle for cancer immunotherapy: AbstractA bottleneck for immunotherapy of cancer is the immunosuppressive microenvironment in which the tumor cells are located. Regardless of the fact that large numbers of tumor-specific T cells can be generated in patients by active immunization or adoptive transfer, these T cells do not readily translate to tumor cell killing in vivo. The immune regulatory mechanism that prevents autoimmunity may be harnessed by tumor cells for the evasion of immune destruction. Regulatory T cells, myeloi...περισσότερα »

Novel window on early human neurodevelopment via fetal exosomes in maternal blood

Αλέξανδρος ΣφακιανάκηςστοMedicine by Alexandros G. SfakianakisΠριν από 9 δευτερόλεπτα
Abstract Adverse in utero exposures can disrupt fetal brain development, deplete subpopulations of neurons and inhibit formation of normal synaptic connections. A major roadblock to unraveling the precise mechanisms and timing of human neurodevelopmental derangement is the almost complete absence of sensitive noninvasive assessments. We present novel methods for isolating fetal neuronal exosomes from maternal plasma as a noninvasive platform for testing aspects of fetal neurodevelopment as early as the 1st trimester. Our methodology represents an important breakthrough both in unde...περισσότερα »

High-throughput sequencing of immune repertoires in multiple sclerosis

Αλέξανδρος ΣφακιανάκηςστοMedicine by Alexandros G. SfakianakisΠριν από 9 δευτερόλεπτα
Abstract T cells and B cells are crucial in the initiation and maintenance of multiple sclerosis (MS), and the activation of these cells is believed to be mediated through specific recognition of antigens by the T- and B-cell receptors. The antigen receptors are highly polymorphic due to recombination (T- and B-cell receptors) and mutation (B-cell receptors) of the encoding genes, which can therefore be used as fingerprints to track individual T- and B-cell clones. Such studies can shed light on mechanisms driving the immune responses and provide new insights into the pathogenesis.... περισσότερα »

The Grooved Rodent Incisor Recapitulates Rudimentary Teeth Characteristics of Ancestral Mammals

Αλέξανδρος ΣφακιανάκηςστοMedicine by Alexandros G. SfakianakisΠριν από 9 δευτερόλεπτα
It is known from the paleontology studies of eutherian mammals that incisor numbers were reduced during evolution. The evolutionary lost incisors may remain as vestigial structures at embryonic stages. The recapitulation of the incisor patterns among mammalian species will potentially uncover the mechanisms underlying the phenotypic transition of incisors during evolution. Here, we showed that a minute tooth formed in the presumptive groove region of the gerbil upper incisor at the early developmental stages, during which multiple epithelial swellings and *Shh* transcription domai... περισσότερα »

Epithelial Microvesicles Promote an Inflammatory Phenotype in Fibroblasts

Αλέξανδρος ΣφακιανάκηςστοMedicine by Alexandros G. SfakianakisΠριν από 9 δευτερόλεπτα
Microvesicles (MVs) are extracellular vesicles secreted by various cell types that are involved in intercellular communication. We hypothesized that in human periodontal disease, the pocket epithelium releases MVs, which then modulate gene expression in the underlying fibroblasts to control periodontal inflammation. MVs were isolated from culture medium of gingival epithelial cells (GECs) treated with oral bacterial biofilm extract or left untreated. Biofilm treatment significantly increased MV release from the GECs. Mass spectrometry of GEC-MVs identified a total of 2,173 protein... περισσότερα »

Thrombin Activates Latent TGF{beta}1 via Integrin {alpha}v{beta}1 in Gingival Fibroblasts

Αλέξανδρος ΣφακιανάκηςστοMedicine by Alexandros G. SfakianakisΠριν από 9 δευτερόλεπτα
Transforming growth factor β (TGFβ) regulates cell proliferation, differentiation, migration, apoptosis, and extracellular matrix production. It also plays a pivotal role in the pathogenesis of gingival overgrowth. Thrombin is a key player in tissue repair, remodeling, and fibrosis after an injury, and it exerts profibrotic effects by activating protease-activated receptors. Connective tissue growth factor (CTGF or CCN2) modulates cell adhesion, migration, proliferation, matrix production, and wound healing. It is overexpressed in many fibrotic disorders, including gingival overgr... περισσότερα »

Enamelin Directs Crystallite Organization at the Enamel-Dentine Junction

Αλέξανδρος ΣφακιανάκηςστοMedicine by Alexandros G. SfakianakisΠριν από 9 δευτερόλεπτα
Enamel is an acellular material formed by the intricate process of amelogenesis. Disruption caused at the initial stages of development, by means of mutations in the *ENAM* gene encoding the enamelin protein, results in enamel hypoplasia. Little is known about the consequence of *ENAM* mutation on the enamel structure at a crystallographic level. The aim of this study was to characterize the structure of *ENAM-*mutated enamel to develop a deeper understanding of the role of enamelin protein during formation with regard to crystal organization. While the activation of Gαi/o-coupled receptors commonly results in postsynaptic responses that show acute desensitization, the presynaptic inhibition of transmitter release caused by many Gαi/o-coupled receptors is maintained during agonist exposure. However, an exception has been noted where GABAB receptor-mediated inhibition of inhibitory postsynaptic currents (IPSCs) recorded in mouse proopiomelanocortin (POMC) neurons exhibit acute desensitization in ~25% of experiments. To determine if differential effector coupling confers sensitivity to desensitization, voltage-clamp recor... περισσότερα »

Voltage-Sensitive Dye (VSD) imaging produces an unprecedented real-time and high-resolution mesoscopic the cortical population activity. We have previously shown that the neuronal compartments contributions to the signal are dynamic and stimulus-dependent (Chemla and Chavane 2010a). Moreover, the VSD signal can also be strongly affected by the network state, such as in anesthetized vs. awake preparations. Here, we investigated the impact of the network state, through GABAA receptors modulation, on the VSD signal using a computational approach. We therefore measured systematically t... περισσότερα »

Sensory-driven behaviors engage a cascade of cortical regions to process sensory input and generate motor output. To investigate the temporal dynamics of neural activity at this global scale, we have developed methods to perform functional imaging across large areas of cortex using a transgenic mouse expressing GCaMP6s, integrated with a head-fixed visual discrimination behavior. This technique allows imaging of activity across the dorsal surface of cortex at a spatial resolution of approximately 50μm and temporal resolution of approximately 100 msec. Auditory Discrimination of Lexical Stress Patterns in Hearing-Impaired Infants with Cochlear Implants Compared with Normal Hearing: Influence of Acoustic Cues and Listening Experience to the Ambient Language

Alexandros G. SfakianakisστοΑκοή και ΙσορροπίαΠριν από 1 ώρα
[image: image]Objectives: To assess discrimination of lexical stress pattern in infants with cochlear implant (CI) compared with infants with normal hearing (NH). While criteria for cochlear implantation have expanded to infants as young as 6 months, little is known regarding infants' processing of suprasegmental-prosodic cues which are known to be important for the first stages of language acquisition. Lexical stress is an example of such a cue, which, in hearing infants, has been shown to assist in segmenting words from fluent speech and in distinguishing between words that differ... περισσότερα »

Intracellular soluble alpha Synuclein oligomers reduce pyramidal cell excitability

Abstract The presynaptic protein alpha-Synuclein (αSyn) aggregates during Parkinson's disease (PD) to form large proteinaceous amyloid plaques, the spread of which throughout the brain clinically defines the severity of the disease. During early stages of aggregation, αSyn forms soluble annular oligomers that show greater toxicity than much larger fibrils. These oligomers produce toxicity through a number of possible mechanisms including the production of pore-forming complexes that permeabilise membranes. In this study, two well-defined species of soluble αSyn oligomers were prod...περισσότερα »

Abstract Spinal Muscular Atrophy (SMA) is a group of autosomal recessive neurodegenerative diseases differing in their clinical outcome, characterized by the specific loss of spinal motor neurons, caused by insufficient level of SMN protein expression. No cure is presently available for SMA. While physical exercise might represent a promising approach for alleviating SMA symptoms, the lack of data dealing with the effects of different exercise types on diseased motor units still precludes the use of active physiotherapy in SMA patients. Quantification of cooperativity in heterodimer-DNA binding improves the accuracy of binding specificity models Computational Biology

Many transcription factors (TFs) have the ability to cooperate on DNA elements as heterodimers. Despite the significance of TF heterodimerization for gene regulation, a quantitative understanding of cooperativity between various TF dimer partners and its impact on heterodimer DNA binding specificity models is still lacking. Here, we used a novel integrative approach, combining microfluidics-steered measurements […]

TORC2-Gad8 is found in the nucleus where it interacts with the MBF transcriptional complex to regulate the response to DNA replication stress. Signal Transduction

The Target of Rapamycin (TOR) kinase is found at the core of two evolutionarily conserved complexes, known as TOR complexes 1 and 2 (TORC1 and TORC2). In fission yeast, TORC2 is dispensable for proliferation under optimal growth conditions but is required for starvation and stress responses. We have previously reported that loss of function of […]

SIRT3 regulates {alpha}-SMA production through the succinate dehydrogenase-GPR91 pathway in hepatic stellate cells Metabolism

Sirtuin 3 (SIRT3) is an NAD+-dependent protein deacetylase. Recent studies have shown that SIRT3 expression is decreased in nonalcoholic fatty liver disease (NAFLD). Moreover, SIRT3 is a key regulator of succinate dehydrogenase (SDH), which catalyzes the oxidation of succinate to fumarate. Increased succinate concentrations and the specific G protein-coupled receptor 91 (GPR91) are involved in […]
