A novel case presenting with an unusual ureteral diverticular lesion similar to adenomyomatous hyperplasia of the gallbladder

Alexandros SfakianakisστοHead and Neck Diseases by Alexandros G.SfakianakisΠριν από 1 λεπτό
We present the first case of an unusual ureteral diverticular lesion demonstrating similarities to adenomyomatous hyperplasia of the gallbladder. A 68-year-old asymptomatic Japanese man with high prostate-specific antigen levels was clinically evaluated. Left hydronephrosis and benign prostatic hyperplasia were detected. A bilateral retrograde pyelogram revealed that the upper and middle portions of the left ureter exhibited an irregular narrow lumen and some pooling of contrast material, which was compatible with ureteral pseudodiverticulosis. Although no malignant cells were see...περισσότερα »

miR-1, regulated by LMP1, suppresses tumour growth and metastasis by targeting K-ras in nasopharyngeal carcinoma

Alexandros SfakianakisστοHead and Neck Diseases by Alexandros G.SfakianakisΠριν από 1 λεπτό
Summary There is evidence to show that downregulation of miR-1 expression is closely related to cancer progression, including in nasopharyngeal carcinoma (NPC). However, the molecular mechanisms underlying miR-1 downregulation in NPC remain largely unknown, especially its association with Epstein–Barr virus (EBV). In this study we found that restoration of miR-1 dramatically inhibited cell invasion *in vitro*, together with tumour growth and metastasis *in vivo*. Importantly, we found that LMP1, an Epstein–Barr virus (EBV)-associated protein, suppressed miR-1 expression. Furthermo... περισσότερα »

Reduced AMPKα2 protein expression restores glucose-induced insulin secretion in islets from calorie-restricted rats

Alexandros SfakianakisστοHead and Neck Diseases by Alexandros G.SfakianakisΠριν από 1 λεπτό
Summary In a state of caloric restriction (CR), improved insulin action was associated with the activation of AMP-activated kinase (AMPK). Here, we verified whether AMPK was involved in impaired β-cell function in islets from rats subjected to CR for 21 days. Eight-week-old male rats were distributed into a control (CTL) group that was fed an isocaloric diet *ad libitum* or a CR group that received 60% of the food consumed by the CTL group. From days 18–21, CTL and CR rats were treated with sense (CTLS and CRS) or antisense (CTLAS and CRAS) AMPKα2 oligonucleotides. Caloric restri... περισσότερα »

Multiple pathways regulate Cten in colorectal cancer without a Tensin switch

Alexandros SfakianakisστοHead and Neck Diseases by Alexandros G.SfakianakisΠριν από 1 λεπτό
Summary *CTEN/TNS4* is a member of the Tensin gene family. It localizes to focal adhesions and induces cell motility. The mechanisms regulating Cten expression are unclear although we have shown regulation by Kras in the colon and pancreas. In normal mammary cell lines, it is reportedly upregulated by epidermal growth factor receptor (EGFR) and STAT3 signalling and upregulation is accompanied by downregulation of Tensin 3 (Tensin switch). In this study, we investigated the roles of EGFR and STAT3 signalling in the regulation of Cten in colorectal cancer (CRC). In addition, we inve... περισσότερα »

Alexandros SfakianakisστοHead and Neck Diseases by Alexandros G.SfakianakisΠριν από 1 λεπτό
Summary Oxidative damage is a central feature of ulcerative colitis. Here, we tested whether the antioxidant Mesna, when administered alone or in combination with n-3 polyunsaturated fatty acids (n-3 PUFAs), affects the outcome of dextran sodium sulphate (DSS)-induced ulcerative colitis in rats. After the induction of colitis, DSS-treated rats were further treated orally (p.o), intraperitoneally (i.p) or intrarectally (i.r) for either 7 or 14 days with Mesna, n-3 PUFAs or both. Rats were euthanized at the end of each treatment period. Clinical disease activity index was recorded t... περισσότερα »

Recombinant human brain natriuretic peptide attenuates trauma-/haemorrhagic shock-induced acute lung injury through inhibiting oxidative stress and the NF-κB-dependent inflammatory/MMP-9 pathway

Summary Acute lung injury (ALI) is one of the most serious complications in traumatic patients and is an important part of multiple organ dysfunction syndrome (MODS). Recombinant human brain natriuretic peptide (rhBNP) is a peptide with a wide range of biological activity. In this study, we investigated local changes in oxidative stress and the NF-κB-dependent matrix metalloproteinase-9 (MMP-9) pathway in rats with trauma/haemorrhagic shock (TH/S)-induced ALI and evaluated the effects of pretreatment with rhBNP. Forty-eight rats were randomly divided into four groups: sham operati... περισσότερα »

Cadmium induces mucin 8 expression via Toll-like receptor 4–mediated extracellular signal related kinase 1/2 and p38 mitogen-activated protein kinase in human airway epithelial cells

Alexandros SfakianakisστοHead and Neck Diseases by Alexandros G.SfakianakisΠριν από 1 λεπτό
Background Inhalation of cadmium can lead to development of inflammatory airway diseases such as acute pulmonary edema and chronic obstructive pulmonary disease. In inflammatory airway diseases, expression of mucins is increased, which leads to increased morbidity and mortality of the affected patients. However, no study on the effect of cadmium on expression of mucin genes in airway epithelial cells has been reported. Therefore, this study was conducted in order to investigate the effect and the brief signaling pathway of cadmium on expression of mucin genes in human airway epithe...περισσότερα »

Mouse is the new woman? Translational research in reproductive immunology

Alexandros SfakianakisστοHead and Neck Diseases by Alexandros G.SfakianakisΠριν από 1 λεπτό
Abstract In an outbred mating typical of human reproduction, the embryo and feto-placental unit express paternal antigens to which the mother's immune system can react. However, the embryo and feto-placental unit can engineer the maternal immune defense system towards helpful rather than harmful reactions. Indeed, this begins with the prospective mother's exposure to paternal seminal plasma. In this review, the pregnancy complications of implantation failure (infertility), recurrent spontaneous abortion, pre-eclampsia and intrauterine growth restriction, and premature labor are ex... περισσότερα »

Caloric restri... περισσότερα »

miR-1, regulated by LMP1, suppresses tumour growth and metastasis by targeting K-ras in nasopharyngeal carcinoma

The purpose of this article is to present the study of a new miniplate designed to keep the maximum strains developed in the cortical surface near the fracture line during accidental biting to values below the threshold causing bone resorption. Designed to offer maximum fracture stability with minimal implanted volume and patient intrusion, the design uses a novel approach to account for the effects of the distance from the fracture line to the nearest screws. Its geometry minimizes the peak forces that can develop during most cases of mandible biomechanical loadings. from #ΓναθοΧ... περισσότερα »

Is p16-positive oropharyngeal squamous cell carcinoma associated with favorable prognosis? A systematic review and meta-analysis

Squamous cell cancers of the head and neck as a group represent the sixth most common malignancy worldwide [1]. While the overall incidence of head and neck cancers (HNC) at various anatomic subsites such as the ventrolateral tongue and floor of mouth has decreased in conjunction with reduced smoking and drinking rates, oropharyngeal squamous cell carcinomas (OPSCC) have increased mainly due to high-risk human papillomavirus (HPV) infection [2]. HPV is a sexually transmitted infection, and currently recognized as the most prevalent sexually transmitted infection in the United Stat...περισσότερα »

Publication date: March 2016 *Source:*International Immunopharmacology, Volume 32 Author(s): Feng Wang, Lie Mao, Hongyan Hou, Shiji Wu, Min Huang, Botao Yin, Jing Huang, Qin Zhu, Yingying Pan, Ziyong Sun Mycobacterium tuberculosis (Mtb)-specific IFN-γ secretion plays important roles in anti-tuberculosis (TB) immunity. Mtb-specific IFN-γ response can be induced in HIV/TB co-infected patients with a low CD4 lymphocyte count; this suggests that the source of Mtb-specific IFN-γ production is not limited in CD4+ T lymphocytes. Currently, the major sources of Mtb-specific IFN-γ production... περισσότερα »

Transgenic mouse lines help decipher the roles of EGFR ligands in the skin

Alexandros G. SfakianakisστοΑισθητική ΔερματολογίαΠριν από 1 ώρα
Abstract The epidermal growth factor receptor (EGFR/ERBB1), a prototypical tyrosine kinase receptor belonging to the ERBB family (ERBB1-ERBB4), has numerous critical roles during development, growth, and tissue homeostasis. Its essentially pro-proliferative actions are frequently exploited by tumor cells, and blocking the activity of the EGFR and of the related ERBB receptors with monoclonal antibodies or kinase inhibitors represents an important strategy for the therapy of various cancer types (1). Canonical activation of the EGFR involves binding by a ligand, induction of recepto... περισσότερα »

In situ chemical behaviour of methylisothiazolinone (MI) and methylchloroisothiazolinone (MCI) in reconstructed human epidermis: a new approach to the cross-reactivity issue

Alexandros G. SfakianakisστοΑισθητική ΔερματολογίαΠριν από 1 ώρα
Summary Background Methylisothiazolinone (MI) [with methylchloroisothiazolinone (MCI) in a ratio of 1:3, a well-recognized allergenic preservative] was released as an individual preservative in the 2000s for industrial products and in 2005 for cosmetics. The high level of exposure to MI since then has provoked an epidemic of contact allergy to MI, and an increase in MI/MCI allergy. There are questions concerning the MI/MCI cross-reaction pattern. Objectives To bring a new perspective on the MI/MCI cross-reactivity issue by studying their *in situ* chemical behaviour in 3D reconst... περισσότερα »

Summary Background In 2003, the EU Cosmetics Directive stated that 26 fragrance substances must be listed on the cosmetic product ingredient labels. Not all of these 26 fragrance substances are detected by the usual screening markers comprising fragrance mix I, fragrance mix II, and *Myroxylon pereirae*. Objectives To evaluate the usefulness of testing with the 26 individual fragrance substances in addition to the standard fragrance screening markers. Materials and Methods Three hundred and twelve consecutive patients were patch tested with our baseline series and the 26 specifi... περισσότερα »

WES in a family trio suggests involvement of TECPR2 in a complex form of progressive motor neuron disease

Alexandros G. SfakianakisστοEvolution of a geneΠριν από 2 ώρες
ABSTRACT We have performed whole exome sequencing in a family trio with a 16 year old girl suffering of progressive motor neuron disease. There was no family history of the disease and no parental consanguinity. Our exome analysis indicated the proband as a compound heterozygote for two missense variants in the TECPR2 gene according to a recessive mode of inheritance. The TECPR2 gene has been reported as a positive regulator of autophagy which is an essential mechanism for maintaining neuron homeostasis and survival and plays a key role in major adult and pediatric neurodegenerativ... περισσότερα »

Reduced AMPKα2 protein expression restores glucose-induced insulin secretion in islets from calorie-restricted rats

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 2 ώρες
Summary In a state of caloric restriction (CR), improved insulin action was associated with the activation of AMP-activated kinase (AMPK). Here, we verified whether AMPK was involved in impaired β-cell function in islets from rats subjected to CR for 21 days. Eight-week-old male rats were distributed into a control (CTL) group that was fed an… Continue reading Reduced AMPKα2 protein expression restores glucose-induced insulin secretion in islets from calorie-restricted rats
