Summary Dermatophytes constitute a complex group of fungi, comprised of by the genera *Trichophyton*, *Epidermophyton* and *Microsporum*. They have the ability to degrade keratin and cause human and animal infections. Molecular techniques have made their identification faster and more accurate, and allowed important advances in phylogenetic studies. We aim to identify molecularly and to determine the phylogenetic relationships in dermatophyte fungi from Brazil and other Latin American countries, using DNA sequencing of the nuclear ribosome regions ITS1-5.8S-ITS2 and D1/D2. Expression of the genes encoding the CasK/R two-component system and the DesA desaturase during Bacillus cereus cold adaptation

Alexandros G. SfakianakisστοΛοίμωξηΠριν από 30 δευτερόλεπτα
Two-component systems (TCS) allow a cell to elaborate a variety of adaptive responses to environment changes. The recently discovered CasK/R TCS plays a role in the optimal unsaturation of fatty acids necessary for cold adaptation of the foodborne pathogen *Bacillus cereus*. Here, we showed that the promoter activity of the operon encoding this TCS was repressed during growth at low temperature in the stationary phase in the parental strain when compared to the *casK/R* mutant, suggesting that CasR negatively regulates the activity of its own promoter in these conditions. Alexandros G. SfakianakisστοΑισθητική ΔερματολογίαΠριν από 30 δευτερόλεπτα
Abstract In the treatment of psoriasis and psoriatic arthritis, recently approved medications undergo the 'early benefit assessment' in Germany. Psoriasis treatments differ in a multiplicity of characteristics like side-effects, beneficial effect, cost and process attributes, which serve to evaluate the patient-relevant benefit compared to standard treatments. Patient preferences might help to aggregate the various patient-relevant outcomes into a single measure. In this context, besides the calculation of the quality-adjusted life-years (QALYs), the Institute for Quality and Effi... περισσότερα »

TRAP1 and SDH as Therapeutic Targets of HCC

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 1 ώρα
S-nitrosoglutathione reductase (GSNOR) represents the best-documented denitrosylase implicated in regulating the levels of proteins posttranslationally modified by nitric oxide on cysteine residues by S-nitrosylation. GSNOR controls a diverse array of physiologic functions, including cellular growth and differentiation, inflammation, and metabolism. Chromosomal deletion of GSNOR results in pathologic protein S-nitrosylation that is implicated in human hepatocellular… Continue reading TRAP1 and SDH as Therapeutic Targets of HCC

Targeting a Glycolysis HIF-1 Feed-Forward Loop

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 1 ώρα
The hypoxia-inducible transcription factor HIF1α drives expression of many glycolytic enzymes. Here, we show that hypoxic glycolysis, in turn, increases HIF1α transcriptional activity and stimulates tumor growth, revealing a novel feed-forward mechanism of glycolysis-HIF1α signaling. Negative regulation of HIF1α by AMPK1 is bypassed in hypoxic cells, due to ATP elevation by increased glycolysis, thereby preventing… Continue reading Targeting a Glycolysis HIF-1 Feed-Forward Loop

Expression of the genes encoding the CasK/R two-component system and the DesA desaturase during Bacillus cereus cold adaptation

Alexandros G. SfakianakisστοΛοίμωξηΠριν από 1 ώρα
This commentary describes an assessment exercise known as the TRIPSE (Tri-Partite Problem Solving Exercise) that mimics science in operation. Students frame hypotheses based on limited data, design experiments to test them, which they later revise with new information. It is emphasised that there are no single correct answers, only sets with varying degrees of plausibility. The approach is flexible and can be adapted to any of the basic biomedical sciences and for students at multiple levels, undergraduate to graduate. Secretion of the powerful angiogenic factor MFG-E8 by pericytes can bypass the therapeutic effects of anti-VEGF therapy, but the mechanisms by which MFG-E8 acts are not fully understood. In this study, we investigated how this factor acts to promote the growth of melanomas that express it. We found that mouse bone marrow–derived mesenchymal stromal cells (MSC) expressed a substantial amount of MFG-E8. To assess its expression from this cell type, we implanted melanoma cells and MSC derived from wild type (WT) or MFG-E8 deficient [knockout (KO)] into mice and monitored tumor growth... περισσότερα »

Targeting a Glycolysis HIF-1 Feed-Forward Loop

The hypoxia-inducible transcription factor HIF1α drives expression of many glycolytic enzymes. Here, we show that hypoxic glycolysis, in turn, increases HIF1α transcriptional activity and stimulates tumor growth, revealing a novel feed-forward mechanism of glycolysis-HIF1α signaling. Negative regulation of HIF1α by AMPK1 is bypassed in hypoxic cells, due to ATP elevation by increased glycolysis, thereby preventing […]

Hydrogen Sulfide Regulates Homeostasis of Mesenchymal Stem Cells and Regulatory T Cells

Alexandros G. SfakianakisστοOroFacial Medicine by Alexandros G.SfakianakisΠριν από 1 ώρα
Hydrogen sulfide (H2S) has long been known as a toxic gas. However, recently accumulated evidence suggests that H2S contributes to a variety of physiologic and pathologic processes. Endogenous H2S production is regulated by multiple enzymes that are differentially expressed in the cardiovascular, neuronal, immune, renal, respiratory, gastrointestinal, reproductive, liver, and endocrine systems. Alteration of H2S metabolism may affect multiple signaling pathways and tissue homeostasis. The growing number of diverse targets for which H2S serves as a gasotransmitter has been extensiv... περισσότερα »

MFG-E8 Promotes MSC-Induced Angiogenesis and TAM in Melanoma: Secretion of the powerful angiogenic factor MFG-E8 by pericytes can bypass the therapeutic effects of anti-VEGF therapy, but the mechanisms by which MFG-E8 acts are not fully understood. In this study, we investigated how this factor acts to promote the growth of melanomas that express it. We… Continue reading MFG-E8 Promotes MSC-Induced Angiogenesis and TAM in Melanoma

Adipocyte Exosomes: A New Link between Obesity and Cancer

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 2 ώρες
Adipocyte Exosomes: A New Link between Obesity and Cancer: Malignant progression results from a dynamic cross-talk between stromal and cancer cells. Recent evidence suggests that this cross-talk is mediated to a significant extent by exosomes, nanovesicles secreted by most cell types and which allow the transfer of proteins, lipids, and nucleic acids between cells. Adipocytes… Continue reading Adipocyte Exosomes: A New Link between Obesity and Cancer

Tissue-resident and memory properties of human T-cell and NK-cell subsets

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 2 ώρες
Efficient immune responses to invading pathogens are the result of the complex but coordinated synergy between a variety of cell types from both the innate and adaptive arms of the immune system. While adaptive and innate immune responses are highly complementary, some cells types within these two systems perform similar functions, underscoring the need for… Continue reading Tissue-resident and memory properties of human T-cell and NK-cell subsets

Targeting a Glycolysis HIF-1 Feed-Forward Loop

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 2 ώρες

TRAP1 and SDH as Therapeutic Targets of HCC

orlsfakianakisστοOtoRhinoLaryngology by Sfakianakis G.AlexandrosΠριν από 2 ώρες
S-nitrosoglutathione reductase (GSNOR) represents the best-documented denitrosylase implicated in regulating the levels of proteins posttranslationally modified by nitric oxide on cysteine residues by S-nitrosylation. GSNOR controls a diverse array of physiologic functions, including cellular growth and differentiation, inflammation, and metabolism. Chromosomal deletion of GSNOR results in pathologic protein S-nitrosylation that is implicated in human hepatocellular… Continue reading TRAP1 and SDH as Therapeutic Targets of HCC

Vascular responsiveness measured by tissue oxygen saturation reperfusion slope is sensitive to different occlusion durations and training status

The reperfusion rate of near-infrared spectroscopy (NIRS)-derived measures of tissue oxygen saturation (StO2) represent vascular responsiveness. This study examined if the reperfusion slope of StO2 is sensitive to different ischemic conditions (i.e., a dose-response) and if differences exist between two groups of different fitness levels. Nine healthy trained (T; Age: 25 ± 3 yr; VO2max: 63.4 ± 6.7 ml kg−1 min−1) and nine healthy untrained (UT; Age: 21 ± 1 yr; VO2max: 46.6 ± 2.5 ml kg−1 min−1) men performed a series of vascular occlusion tests of different durations (30 s, 1 min, ... περισσότερα »
