Rapamycin Dosage

The mTOR pathway is a critical regulator of cell growth, proliferation, metabolism, and survival. Dysregulation of mTOR signaling has been observed in most cancers and, thus, the mTOR pathway has been extensively studied for therapeutic intervention. Rapamycin is a natural product that inhibits mTOR with high specificity. However, its efficacy varies by dose in several […]

HSP90 Inhibition Kills Mutant KRAS Colon Cancer Cells

Oncogenic mutations of KRAS pose a great challenge in the treatment of colorectal cancer. Here we report that mutant KRAS colon cancer cells are nevertheless more susceptible to apoptosis induced by the HSP90 inhibitor AUY922 than those carrying wild-type KRAS. Although AUY922 inhibited HSP90 activity with comparable potency in colon cancer cells irrespective of their […]

HMGB1 Predicts Radiotherapy Response in Bladder Cancer

Although radical cystectomy surgery is the standard-of-care for muscle-invasive bladder cancer, it entails complete removal of the bladder and surrounding organs which leads to substantial loss in the quality-of-life of patients. Radiotherapy, which spares the bladder, would be a more appropriate treatment modality if we can utilize molecular markers to select patients with better response […]

WIP1 Inhibitor GSK2830371 Potentiates Nutlin-3/RG7388 Sensitivity

Sensitivity to MDM2 inhibitors is widely different among responsive TP53 wild-type cell lines and tumors. Understanding the determinants of MDM2 inhibitor sensitivity is pertinent for their optimal clinical application. Wild-type p53-inducible phosphatase-1 (WIP1) encoded by PPM1D, is activated, gained/amplified in a range of TP53 wild-type malignancies, and is involved in p53 stress response homeostasis. We […]

Vaccine Targeting of Subdominant CD8+ T Cell Epitopes Increases the Breadth of the T Cell Response upon Viral Challenge, but May Impair Immediate Virus Control [IMMUNOTHERAPY AND VACCINES]: As a result of the difficulties in making efficient vaccines against genetically unstable viruses such as HIV, it has been suggested that future vaccines should preferentially target subdominant epitopes, the idea being that this should allow a greater breadth of the induced T cell response and, hence, a greater efficiency in controlling escape variants. However, to our knowledge the evidence su...περισσότερα »

Combined magnetic fields provide robust coverage for interbody and posterolateral lumbar spinal fusion sites

Alexandros G. SfakianakisστοΒιοΜηχανικήΠριν από 41 δευτερόλεπτα
Abstract Electromagnetic fields generated by spinal bone growth stimulation devices have been computationally modelled to determine coverage of the lumbar spinal vertebrae. The underlying assumption of these models was that the electric field, but not the magnetic field, was therapeutically relevant. However, there are no published studies examining the therapeutic coverage of spinal fusion sites by stimulators utilizing combined magnetic fields. To assess the coverage, an anatomical model of the vertebrae and discs of the lumbar spine was developed to represent interbody and poste... περισσότερα »

A Tumor-Selective Picornavirus Promotes Tumor Regression

Alexandros G. SfakianakisστοΟγκολογίαΠριν από 53 δευτερόλεπτα
Picornaviruses have emerged as promising cancer therapies due to their ability to drive cytotoxic cellular immune responses and for promoting oncolysis. These properties include preferential replication in tumor cells, the induction of strong innate and adaptive immune responses, and the ease with which their genomes can be manipulated. We have developed Theiler's murine encephalomyelitis virus (TMEV) as an immunotherapy vector that promotes strong adaptive immune responses to tumor antigens embedded within its genome. To further explore its usefulness as cancer therapy, we invest... περισσότερα »

While pharmacoepidemiologic and laboratory studies have supported the hypothesis that the antidiabetic drug metformin may be useful in treating or preventing cancer, there is limited evidence to suggest which specific cancer sites may be particularly sensitive. Sensitivity likely is determined both by features of tumor pathophysiology and by pharmacokinetic factors. We used UPII-mutant Ha-ras transgenic… Continue reading Metformin Inhibits Urothelial Tumorigenesis

Does a coupling capacitor enhance the charge balance during neural stimulation? An empirical study

Alexandros G. SfakianakisστοΒιοΜηχανικήΠριν από 57 δευτερόλεπτα
Abstract Due to their DC-blocking characteristic, coupling capacitors are widely used to prevent potentially harmful charge buildup at the electrode–tissue interface. Although the capacitors can be an effective safety measure, it often seems overlooked that coupling capacitors actually introduce an offset voltage over the electrode–tissue interface as well. This work investigates this offset voltage both analytically and experimentally. The calculations as well as the experiments using bipolar-driven platinum electrodes in a saline solution confirm that coupling capacitors introduc... περισσότερα »

Effects of reconstructed magnetic field from sparse noisy boundary measurements on localization of active neural source

Alexandros G. SfakianakisστοΒιοΜηχανικήΠριν από 57 δευτερόλεπτα
Abstract Localization of active neural source (ANS) from measurements on head surface is vital in magnetoencephalography. As neuron-generated magnetic fields are extremely weak, significant uncertainties caused by stochastic measurement interference complicate its localization. This paper presents a novel computational method based on reconstructed magnetic field from sparse noisy measurements for enhanced ANS localization by suppressing effects of unrelated noise. In this approach, the magnetic flux density (MFD) in the nearby current-free space outside the head is reconstructed ... περισσότερα »

In the current study, the potential ISO inhibition of bladder tumor formation […]

CAR Agonist Improves CHOP-Based Chemotherapy

The constitutive androstane receptor (CAR and NR1i3) is a key regulator of CYP2B6, the enzyme predominantly responsible for the biotransformation of cyclophosphamide (CPA) to its pharmacologically active metabolite, 4-hydroxycyclophosphamide (4-OH-CPA). Previous studies from our laboratory illustrated that CAR activation increases the formation of 4-OH-CPA; however, CPA is rarely used clinically outside of combination therapies. Here, […]

Thymus medulla under construction: Time and Space Oddities

Αλέξανδρος ΣφακιανάκηςστοOtoRhinoLaryngology by Alexandros G.SfakianakisΠριν από 1 ώρα
The development of effective T-cell-based immunotherapies to treat infection, cancer and autoimmunity should incorporate the ground rules that control differentiation of T cells in the thymus. Within the thymus, thymic epithelial cells (TECs) provide microenvironments supportive of the generation and selection of T cells that are responsive to pathogen-derived antigens, and yet tolerant to self-determinants. Defects in TEC differentiation cause syndromes that range from immunodeficiency to autoimmunity, which makes the study of TECs of fundamental and clinical importance to compr... περισσότερα »
