Correlations between prefrontal neurons form a small-world network that optimizes the generation of multineuron sequences of activity

Alexandros G. SfakianakisστοPhysiology by Alexandros G.SfakianakisΠριν από 7 δευτερόλεπτα
Sequential patterns of prefrontal activity are believed to mediate important behaviors, e.g., working memory, but it remains unclear exactly how they are generated. In accordance with previous studies of cortical circuits, we found that prefrontal microcircuits in young adult mice spontaneously generate many more stereotyped sequences of activity than expected by chance. However, the key question of whether these sequences depend on a specific functional organization within the cortical microcircuit, or emerge simply as a by-product of random interactions between neurons, remains ... περισσότερα »

Properties and physiological function of Ca2+-dependent K+ currents in uniglomerular olfactory projection neurons

Alexandros G. SfakianakisστοPhysiology by Alexandros G.SfakianakisΠριν από 7 δευτερόλεπτα
Ca2+-activated potassium currents [*I*K(Ca)] are an important link between the intracellular signaling system and the membrane potential, which shapes intrinsic electrophysiological properties. To better understand the ionic mechanisms that mediate intrinsic firing properties of olfactory uniglomerular projection neurons (uPNs), we used whole cell patch-clamp recordings in an intact adult brain preparation of the male cockroach *Periplaneta americana* to analyze *I*K(Ca). In the insect brain, uPNs form the principal pathway from the antennal lobe to the protocerebrum, where center... περισσότερα »

Roles of specific Kv channel types in repolarization of the action potential in genetically identified subclasses of pyramidal neurons in mouse neocortex

Alexandros G. SfakianakisστοPhysiology by Alexandros G.SfakianakisΠριν από 7 δευτερόλεπτα
The action potential (AP) is a fundamental feature of excitable cells that serves as the basis for long-distance signaling in the nervous system. There is considerable diversity in the appearance of APs and the underlying repolarization mechanisms in different neuronal types (reviewed in Bean BP. *Nat Rev Neurosci* 8: 451–465, 2007), including among pyramidal cell subtypes. In the present work, we used specific pharmacological blockers to test for contributions of Kv1, Kv2, or Kv4 channels to repolarization of single APs in two genetically defined subpopulations of pyramidal cells ... περισσότερα »

Blocking trial-by-trial error correction does not interfere with motor learning in human walking

Alexandros G. SfakianakisστοPhysiology by Alexandros G.SfakianakisΠριν από 7 δευτερόλεπτα
Movements can be learned implicitly in response to new environmental demands or explicitly through instruction and strategy. The former is often studied in an environment that perturbs movement so that people learn to correct the errors and store a new motor pattern. Here, we demonstrate in human walking that implicit learning of foot placement occurs even when an explicit strategy is used to block changes in foot placement during the learning process. We studied people learning a new walking pattern on a split-belt treadmill with and without an explicit strategy through instructi... περισσότερα »

An asymmetric outer retinal response to drifting sawtooth gratings

Alexandros G. SfakianakisστοPhysiology by Alexandros G.SfakianakisΠριν από 7 δευτερόλεπτα
Electroretinogram (ERG) studies have demonstrated that the retinal response to temporally modulated fast-ON and fast-OFF sawtooth flicker is asymmetric. The response to spatiotemporal sawtooth stimuli has not yet been investigated. Perceptually, such drifting gratings or diamond plaids shaded in a sawtooth pattern appear brighter when movement produces fast-OFF relative to fast-ON luminance profiles. The neural origins of this illusion remain unclear (although a retinal basis has been suggested). Thus we presented toad eyecups with sequential epochs of sawtooth, sine-wave, and squ...περισσότερα »

Classification of odorants across layers in locust olfactory pathway

Alexandros G. SfakianakisστοPhysiology by Alexandros G.SfakianakisΠριν από 7 δευτερόλεπτα
Olfactory processing takes place across multiple layers of neurons from the transduction of odorants in the periphery, to odor quality processing, learning, and decision making in higher olfactory structures. In insects, projection neurons (PNs) in the antennal lobe send odor information to the Kenyon cells (KCs) of the mushroom bodies and lateral horn neurons (LHNs). To examine the odor information content in different structures of the insect brain, antennal lobe, mushroom bodies and lateral horn, we designed a model of the olfactory network based on electrophysiological recordin... περισσότερα »

Uterine morphology during diapause and early pregnancy in the tammar wallaby (Macropus eugenii)

Abstract In mammals, embryonic diapause, or suspension of embryonic development, occurs when embryos at the blastocyst stage are arrested in growth and metabolism. In the tammar wallaby (Macropus eugenii), there are two separate uteri, only one of which becomes gravid with the single conceptus at a post-partum oestrus, so changes during pregnancy can be compared […]

Over the last decades, around the world, the concept of competitiveness was long debated by economists (and others), widely used and even sometimes overused. Although at the theoretical level, a number of determinant factors of health care organizations' competitiveness have been proposed, their diversity and the little empirical data available argues for the need to create and validate a model of competitiveness of health organizations. The purpose of this paper is (considering the theoretical approach) to shape a model of sustainable competitiveness of health organizations. In t... περισσότερα »

Remodeling of Dendritic Spines in the Avian Vocal Motor Cortex Following Deafening Depends on the Basal Ganglia Circuit

Alexandros G. SfakianakisστοΝευρολογία κ' ΝευροχειρουργικήΠριν από 16 δευτερόλεπτα
Deafening elicits a deterioration of learned vocalization, in both humans and songbirds. In songbirds, learned vocal plasticity has been shown to depend on the basal ganglia-cortical circuit, but the underlying cellular basis remains to be clarified. Using confocal imaging and electron microscopy, we examined the effect of deafening on dendritic spines in avian vocal motor cortex, the robust nucleus of the arcopallium (RA), and investigated the role of the basal ganglia circuit in motor cortex plasticity. We found rapid structural changes to RA dendritic spines in response to hear... περισσότερα »

New Role of ATM in Controlling GABAergic Tone During Development

Alexandros G. SfakianakisστοΝευρολογία κ' ΝευροχειρουργικήΠριν από 16 δευτερόλεπτα
The capacity to guarantee the proper excitatory/inhibitory balance is one of the most critical steps during early development responsible for the correct brain organization, function, and plasticity. GABAergic neurons guide this process leading to the right structural organization, brain circuitry, and neuronal firing. Here, we identified the ataxia telangiectasia mutated (ATM), a serine/threonine protein kinase linked to DNA damage response, as crucial in regulating neurotransmission. We found that reduced levels of ATM in the hippocampal neuronal cultures produce an excitatory/i... περισσότερα »

The Addiction-Related Protein ANKK1 is Differentially Expressed During the Cell Cycle in Neural Precursors

Alexandros G. SfakianakisστοΝευρολογία κ' ΝευροχειρουργικήΠριν από 16 δευτερόλεπτα
*Taq*IA is a polymorphism associated with addictions and dopamine-related traits. It is located in the ankyrin repeat and kinase domain containing 1 gene (*ANKK1*) nearby the gene for the dopamine D2 receptor (D2R). Since ANKK1 function is unknown, *Taq*IA-associated traits have been explained only by differences in D2R. Here we report *ANKK1* studies in mouse and human brain using quantitative real-time PCR, Western blot, immunohistochemistry, and flow cytometry. *ANKK1* mRNA and protein isoforms vary along neurodevelopment in the human and mouse brain. In mouse adult brain ANKK1... περισσότερα »

Look Hear! The Prefrontal Cortex is Stratified by Modality of Sensory Input During Multisensory Cognitive Control

Parsing multisensory information from a complex external environment is a fundamental skill for all organisms. However, different organizational schemes currently exist for how multisensory information is processed in human (supramodal; organized by cognitive demands) versus primate (organized by modality/cognitive demands) lateral prefrontal cortex (LPFC). Functional magnetic resonance imaging results from a large cohort of healthy controls (*N* = 64; Experiment 1) revealed a rostral-caudal stratification of LPFC for auditory versus visual attention during an audio-visual Stroop ... περισσότερα »

Modulation of Functional Connectivity in Auditory-Motor Networks in Musicians Compared with Nonmusicians

Correlation of spontaneous fluctuations at rest between anatomically distinct brain areas are proposed to reflect the profile of individual a priori cognitive biases, coded as synaptic efficacies in cortical networks. Here, we investigate functional connectivity at rest (rs-FC) in musicians and nonmusicians to test for differences in auditory, motor, and audiomotor connectivity. As expected, musicians had stronger rs-FC between the right auditory cortex (AC) and the right ventral premotor cortex than nonmusicians, and this stronger rs-FC was greater in musicians with more years of... περισσότερα »

Nogo-66 Restricts Synaptic Strengthening via Lingo1 and the ROCK2-Cofilin Pathway to Control Actin Dynamics

Alexandros G. SfakianakisστοΝευρολογία κ' ΝευροχειρουργικήΠριν από 16 δευτερόλεπτα
Nogo-A restricts long-term potentiation (LTP) at the Schaffer collateral–CA1 pathway in the adult hippocampus via 2 extracellular domains: Nogo-A-20 and Nogo-66. Nogo-66 signals via Nogo Receptor 1 (NgR1) to regulate synaptic function. Whether the NgR1 coreceptors Lingo1 and p75 NTR are involved in the signaling in this context is still not known. Moreover, the intracellular cascade mediating the activity of Nogo-66 in restricting LTP is unexplored. We combine electrophysiology and biochemistry in acute hippocampal slices and demonstrate that a loss of function for Lingo1 results i... περισσότερα »

Learned Value Shapes Responses to Objects in Frontal and Ventral Stream Networks in Macaque Monkeys

Alexandros G. SfakianakisστοΝευρολογία κ' ΝευροχειρουργικήΠριν από 16 δευτερόλεπτα
We have an incomplete picture of how the brain links object representations to reward value, and how this information is stored and later retrieved. The orbitofrontal cortex (OFC), medial frontal cortex (MFC), and ventrolateral prefrontal cortex (VLPFC), together with the amygdala, are thought to play key roles in these processes. There is an apparent discrepancy, however, regarding frontal areas thought to encode value in macaque monkeys versus humans. To address this issue, we used fMRI in macaque monkeys to localize brain areas encoding recently learned image values. Each week,... περισσότερα »

Pediatric airway management is a low-frequency, high-risk skill that requires ongoing training and practice to maintain proficiency. Providers may consider utilizing an alternative airway, such as the LMA or the King LT-D, when managing the pediatric airway rather than resorting to intubation. For services that utilize surgical airways for pediatric patients, this skill requires a great deal of ongoing training and practice as evidenced by the poor success rate among many emergency providers. The cornerstone of pediatric airway management remains basic airway maneuvers, and providers should only consider moving to advanced procedures if the basic maneuvers are not effective in maintaining adequate ventilation and oxygenation.

Pediatric Airway Management : Respiratory arrest is the predominant cause of mortality in critically ill and injured children. Prehospital airway management can be life saving for critically ill or injured children but is a low-frequency, high-risk event fraught with the potential for error. Basic airway maneuvers are the cornerstone of maintenance of oxygenation and ventilation by EMS professionals in the field. Overall, the incorporation of pediatric intubation in prehospital protocols has demonstrated more negative than positive impact. In a landmark study comparing prehospital intubation to BVM only, "no statistically significant difference was seen for either survival to hospital discharge or good neurologic outcomes.

Nitrogen (N) and phosphorus (P) cycles are absolutely vital in maintaining sustainable food systems. Human activities disturb the natural balance of these cycles by creating enormous additional nutrient fluxes, causing eutrophication of waterways and pollution in land systems. To tackle this problem, sustainable nutrient management is required. This paper addresses sustainable nutrient management in two countries: The Netherlands and Finland. We adopt a critical perspective on resource politics, especially towards opportunistic policy strategies for the pollutant management of N a... περισσότερα »

We address the problem of bootstrapping security and privacy policies for newly-deployed apps in wireless body area networks (WBAN) composed of smartphones, sensors and other wearable devices. We introduce a framework to model such a WBAN as an undirected graph whose vertices correspond to devices, apps and app resources, while edges model structural relationships among them. This graph is then augmented with attributes capturing the features of each entity together with user-defined tags. We then adapt available graph-based similarity metrics to find the closest app to a new one ... περισσότερα »

Roles of specific Kv channel types in repolarization of the action potential in genetically identified subclasses of pyramidal neurons in mouse neocortex

Association of Genetic Risk for Schizophrenia With Nonparticipation Over Time in a Population-Based Cohort Study

Alexandros G. SfakianakisστοΟγκολογίαΠριν από 1 ώρα
Progress has recently been made in understanding the genetic basis of schizophrenia and other psychiatric disorders. Longitudinal studies are complicated by participant dropout, which could be related to the presence of psychiatric problems and associated genetic risk. We tested whether common genetic variants implicated in schizophrenia were associated with study nonparticipation among 7,867 children and 7,850 mothers from the Avon Longitudinal Study of Parents and Children (ALSPAC; 1991–2007), a longitudinal population cohort study. Higher polygenic risk scores for schizophrenia...περισσότερα »

Coastal erosion as a source of mercury into the marine environment along the Polish Baltic shore

Alexandros G. SfakianakisστοΑισθητική ΔερματολογίαΠριν από 2 ώρες
Abstract The climate changes in recent years in the southern Baltic have been resulting in an increased frequency of natural extreme phenomena (i.e. storms, floods) and intensification of abrasion processes, which leads to introduction of large amounts of sedimentary deposits into the marine environment. The aim of this study was to determine the mercury load introduced to the Baltic Sea with deposits crumbling off the cliffs—parts of the coast that are the most exposed to abrasion. The studies were carried out close to five cliffs located on the Polish coast in the years 2011–201... περισσότερα »
