Cbx2 targets PRC1 to constitutive heterochromatin in mouse zygotes in a parent-of-origin-dependent manner

Polycomb repressive complexes PRC1 and PRC2 regulate expression of genes involved in proliferation and development. In mouse early embryos, however, canonical PRC1 localizes to paternal pericentric heterochromatin (pat-PCH), where it represses transcription of major satellite repeats. In contrast, maternal PCH (mat-PCH) is enriched for H3 lysine 9 tri-methylation (H3K9me3) and Hp1β. How PRC1 is targeted […]

Incorporating Functional Genomic Information in Genetic Association Studies Using an Empirical Bayes Approach

Alexandros SfakianakisστοHead and Neck Diseases by Alexandros G.SfakianakisΠριν από 8 δευτερόλεπτα
ABSTRACT There is a large amount of functional genetic data available, which can be used to inform fine-mapping association studies (in diseases with well-characterised disease pathways). Single nucleotide polymorphism (SNP) prioritization via Bayes factors is attractive because prior information can inform the effect size or the prior probability of causal association. This approach requires the specification of the effect size. If the information needed to estimate a priori the probability density for the effect sizes for causal SNPs in a genomic region isn't consistent or isn't ... περισσότερα »

Cochlear implantation was introduced in the United States as a viable treatment for hearing loss in the 1980s. Originally, the anticipated benefits were awareness of sound and enhancement of speech reading. Therefore, the early candidacy guidelines were very strict. There were concerns that the benefit from a cochlear implant would be limited or potentially be very difficult to measure, so the industry was conservative about who it considered for an implant. Abstract Converging evidence points to glycogen synthase kinase (GSK) 3 as a key player in the pathogenesis of podocytopathy and proteinuria. However, it remains unclear if GSK3 is involved in podocyte autonomous injury in glomerular disease. In normal kidneys, the β isoform of GSK3 was found to be the major GSK3 expressed in glomeruli and intensely stained in podocytes. GSK3β expression in podocytes was markedly elevated in experimental or human proteinuric glomerulopathy. Podocyte specific somatic ablation of GSK3β in adult mice attenuated proteinuria and ameliorated podocyte in... περισσότερα »

Abstract Colorectal cancer (CRC) is one of the most common malignancies and is the second leading cause of cancer death in humans. Tumour suppressor candidate 3 (*TUSC3*) plays an important role in embryogenesis and metabolism. Deletion of *TUSC3* often causes non-syndromic mental retardation. Even though *TUSC3* deregulation is frequently observed in epithelial cancers, the function of *TUSC3* in CRC has remained unknown. In this study, we observed greater expression of TUSC3 at the mRNA and protein level in clinical colorectal tumour samples compared with paired normal tissues. ... περισσότερα »

Methods: In this retrospective analysis, patients with diabetes with microvascular palsies were assessed for the prevalence of diabetic retinopathy (DR). We compared these data with the prevalence of... περισσότερα »

Incorporating Functional Genomic Information in Genetic Association Studies Using an Empirical Bayes Approach

Alexandros SfakianakisστοHead and Neck Diseases by Alexandros G.SfakianakisΠριν από 9 δευτερόλεπτα

Application of Deuteporfin in the Metastatic Lymph Node Mapping of Pancreatic Cancer: An in vivo Study

Alexandros G. SfakianakisστοΑισθητική ΔερματολογίαΠριν από 11 δευτερόλεπτα
Abstract For most cancer patients, the presence of metastatic lymph nodes usually indicates regional recurrence and poor prognosis. Therefore, lymph node mapping is a requisite for disease staging, prognosis prediction, and decision making in the treatment of cancer. Deuteporfin, a second-generation photosensitizer, has a maximum excitation wavelength that can reach the near infrared (NIR) region (650-700 nm). We aimed to take advantage of these aspects of deuteporfin and use it as a fluorescent probe for metastatic lymph node mapping *in vivo* using NIR fluorescent imaging. In ou... περισσότερα »

Perceptual and Brain Response to Odors Is Associated with Body Mass Index and Postprandial Total Ghrelin Reactivity to a Meal: Animal studies have shown that olfactory sensitivity is greater when fasted than when fed. However, human research has generated inconsistent results. One possible explanation for these conflicting findings is metabolic health. Many metabolic peptides, including ghrelin, are moderated by adiposity and influence olfaction and olfactory-guided behaviors. We tested whether the effect of a meal on the perceived intensity of suprathreshold chemosensory stimuli i... περισσότερα »

Abstract Papillary thyroid carcinoma (PTC) is the most common adult thyroid malignancy and often presents with multiple anatomically distinct foci within the thyroid, known as multifocal papillary thyroid carcinoma (MPTC). The widespread application of the next-generation sequencing technologies in cancer genomics research provides novel insights into determining the clonal relationship between multiple tumours within the same thyroid gland. For eight MPTC patients, we performed whole-exome sequencing and targeted region sequencing to identify the non-synonymous point mutations and ...περισσότερα »

TUSC3 promotes colorectal cancer progression and epithelial-mesenchymal transition (EMT) through WNT/β-catenin and MAPK signalling

Αλέξανδρος ΣφακιανάκηςστοMedicine by Alexandros G. SfakianakisΠριν από 50 δευτερόλεπτα
G... περισσότερα »

Podocyte specific somatic ablation of GSK3β in adult mice attenuated proteinuria and ameliorated podocyte inj... περισσότερα »

Large Genomic Rearrangement in Pancreatic Ductal Tumors

Αλέξανδρος ΣφακιανάκηςστοMedicine by Sfakianakis G. AlexandrosΠριν από 1 ώρα
Many somatic mutations have been detected in pancreatic ductal adenocarcinoma (PDAC), leading to the identification of some key drivers of disease progression, but the involvement of large genomic rearrangements has often been overlooked. In this study, we performed mate pair sequencing (MPseq) on genomic DNA from 24 PDAC tumors, including 15 laser-captured microdissected PDAC and 9 patient-derived xenografts, to identify genome-wide rearrangements. Large genomic rearrangements with intragenic breakpoints altering key regulatory genes involved in PDAC progression were detected in ...περισσότερα »

Microscale distribution of virioplankton and heterotrophic bacteria in the Bohai Sea

In light of limited research in the relationship between the microscale distribution and dynamic changes of microplankton in the shallow Bohai Sea, here, we used flow cytometry to analyze samples collected from the Bohai Sea channel in winter and summer. Results showed that both the average of viral abundance (VA) and bacterial abundance (BA) were lower in winter (3.61 x 107 cells/mL and 1.84 x 106 cells/mL, respectively) than in summer (7.47 x 107 cells/mL and 5.05 x 106 cells/mL, respectively). At all 16 stations, VA was one order of magnitude greater than BA, with a positive re... περισσότερα »

Antiangiogenesis Restores Bone Marrow Vascular Niche

Αλέξανδρος ΣφακιανάκηςστοMedicine by Sfakianakis G. AlexandrosΠριν από 1 ώρα
Antiangiogenesis–based cancer therapies, specifically those targeting the VEGF-A/VEGFR2 pathway, have been approved for subsets of solid tumors. However, these therapies result in an increase in hematologic adverse events. We surmised that both the bone marrow vasculature and VEGF receptor–positive hematopoietic cells could be impacted by VEGF pathway–targeted therapies. We used a mouse model of spontaneous breast cancer to decipher the mechanism by which VEGF pathway inhibition alters hematopoiesis. Tumor-bearing animals, while exhibiting increased angiogenesis at the primary tum... περισσότερα »

Oncogenic KRAS Modulates EZH2 Expression in NSCLC

Αλέξανδρος ΣφακιανάκηςστοMedicine by Sfakianakis G. AlexandrosΠριν από 1 ώρα
EZH2 overexpression promotes cancer by increasing histone methylation to silence tumor suppressor genes, but how EZH2 levels become elevated in cancer is not understood. In this study, we investigated the mechanisms by which EZH2 expression is regulated in non–small cell lung carcinoma cells by oncogenic KRAS. In cells harboring KRASG12C and KRASG12D mutations, EZH2 expression was modulated by MEK–ERK and PI3K/AKT signaling, respectively. Accordingly, MEK–ERK depletion decreased EZH2 expression in cells harboring the KRASG12C mutation, whereas PI3K/AKT depletion decreased EZH2 exp... περισσότερα »

DR5 Targeting Triggers Gastrointestinal Toxicity

Αλέξανδρος ΣφακιανάκηςστοMedicine by Sfakianakis G. AlexandrosΠριν από 1 ώρα
The combination of TRAIL death receptor agonists and radiochemotherapy to treat advanced cancers continues to be investigated in clinical trials. We previously showed that normal cells with a functional DNA damage response (DDR) upregulate the expression of death-inducing receptor DR5/TRAILR2/TNFRSF10B in a p53-dependent manner that sensitizes them to treatment with DR5 agonists. However, it is unclear if targeting DR5 selectively sensitizes cancer cells to agonist treatment following exposure to DNA-damaging chemotherapy, and to what extent normal tissues are targeted. Here, we s...περισσότερα »

Inflammation in gastric cancer: Interplay of the COX-2/PGE2 and TLR/MyD88 pathways

Alexandros G. SfakianakisστοΟγκολογίαΠριν από 1 ώρα
Abstract Cyclooxygenase 2 (COX-2) and its downstream product prostaglandin E2 (PGE2) play a key role in generation of inflammatory microenvironment in tumor tissues. Gastric cancer is closely associated with *Helicobacter pylori* infection, which stimulates innate immune responses through Toll-like receptors (TLRs), inducing COX-2/PGE2 pathway through NF-κB activation. A pathway analysis of human gastric cancer shows that both COX-2 pathway and Wnt/β-catenin signaling are significantly activated in tubular-type gastric cancer, while basal levels of these pathways are also increased... περισσότερα »

Sugarcane, Saccharum spp., in the United States is attacked by a number of different arthropod pests. The most serious among those pests are two stalk boring moths in the Family Crambidae: the sugarcane borer, Diatraea saccharalis (F.), and the Mexican rice borer, Eoreuma loftini (Dyar). The two species are affected by abiotic and biotic environmental stress factors. Water deficit and excessive soil nitrogen alter physical and physiochemical aspects of the sugarcane plant that make the crop increasingly vulnerable to E. loftini. Weed growth can be competitive with sugarcane but it... περισσότερα »

Abstract A common strategy when searching for cognitive enhancing drugs has been to target the NMDA receptor, given its putative role in synaptic plasticity and learning. Evidence in favour of this approach has come primarily from studies with rodents using behavioural assays like the Morris watermaze. D-amino acid oxidase (DAO) degrades neutral D-amino acids such as D-serine, the primary endogenous co-agonist acting at the glycine site of the synaptic NMDA receptor. Inhibiting DAO could therefore provide an effective and viable means of enhancing cognition, particularly in disorde... περισσότερα »

An IL-1β-neutralizing or a control antibody was administered intraperitoneally at 30 min following central fluid percussion injury (cFPI), a mouse model of widespread TAI. Animals subjected t... περισσότερα »

Subcellular calcium dynamics during juvenile development in mouse hippocampal astrocytes

Alexandros G. SfakianakisστοΝευρολογία κ' ΝευροχειρουργικήΠριν από 1 ώρα
Abstract Astrocytes generate calcium signals throughout their fine processes, which are assumed to locally regulate neighboring neurotransmission and blood flow. The intercellular morphological relationships mature during juvenile periods when astrocytes elongate highly ramified processes. In this study, we examined developmental changes in calcium activity patterns of single hippocampal astrocytes using a transgenic mouse line in which astrocytes selectively express a genetically encoded calcium indicator, Yellow Cameleon-Nano50. Compared with postnatal day 7, astrocytes at postna...περισσότερα »

Oscillations in the embryonic chick olfactory bulb: initial expression and development revealed by optical imaging with a voltage-sensitive dye

Alexandros G. SfakianakisστοΝευρολογία κ' ΝευροχειρουργικήΠριν από 1 ώρα
Abstract In our previous study, we applied a multiple-site optical recording technique with a voltage-sensitive dye to the embryonic chick olfactory system and showed that functional synaptic transmission in the olfactory bulb was expressed at the embryonic 6-7-day stage. It is known that oscillations, i.e., stereotyped sinusoidal neural activity, appear in the olfactory system of various species. The focus of the present study is to determine whether the oscillation is also generated in the embryonic chick olfactory bulb and, if this is the case, when the oscillation appears and ... περισσότερα »

Striatonigral control of movement velocity in mice

Alexandros G. SfakianakisστοΝευρολογία κ' ΝευροχειρουργικήΠριν από 1 ώρα
Abstract The basal ganglia have long been implicated in action initiation. Using three-dimensional motion capture, we quantified the effects of optogenetic stimulation of the striatonigral (direct) pathway on movement kinematics. We generated transgenic mice with Channelrhodopsin-2 expression in striatal neurons that express the D1-like dopamine receptor. With optic fibers placed in the sensorimotor striatum, an area known to contain movement velocity-related single units, photo-stimulation reliably produced movements that could be precisely quantified with our motion capture prog... περισσότερα »

Prosigna ROR Score Predicts Chemosensitivity

Alexandros G. SfakianakisστοΟγκολογίαΠριν από 2 ώρες
*Purpose:* Most hormone receptor (HR)+/HER2– breast cancer patients respond unfavorably to neoadjuvant chemotherapy (NAC); however, genomic tests may identify those patients who are likely to benefit. Using the Prosigna assay, we first evaluated the technical performance of core needle biopsy (CNB) tissues. We then determined whether Prosigna risk of relapse (ROR) score and intrinsic subtype predicted response to NAC in HR+/HER2– patients using CNB samples. *Experimental Design:* Using the NanoString's nCounter Dx analysis system and a development tissue sample set, we established... περισσότερα »

Therapeutic Wnt Pathway Inhibition in Colorectal Cancer

Alexandros G. SfakianakisστοΟγκολογίαΠριν από 2 ώρες
*Purpose:* Oncogenic mutations in the KRAS/PI3K/AKT pathway are one of the most frequent alterations in cancer. Although PI3K or AKT inhibitors show promising results in clinical trials, drug resistance frequently emerges. We previously revealed Wnt/β-catenin signaling hyperactivation as responsible for such resistance in colorectal cancer. Here we investigate Wnt-mediated resistance in patients treated with PI3K or AKT inhibitors in clinical trials and evaluate the efficacy of a new Wnt/tankyrase inhibitor, NVP-TNKS656, to overcome such resistance. *Experimental Design:* Colorect... περισσότερα »

Prognostic Value of BRAF Mutation Detection in Circulation

Alexandros G. SfakianakisστοΟγκολογίαΠριν από 2 ώρες
*Purpose:* Tumor-derived circulating cell–free DNA (cfDNA) is a potential alternative source from which to derive tumor mutation status. cfDNA data from four clinical studies of the BRAF inhibitor (BRAFi) dabrafenib or the MEK inhibitor (MEKi) trametinib were analyzed to determine the association between *BRAF* mutation status in cfDNA and tumor tissue, and the association of *BRAF* cfDNA mutation status with baseline factors and clinical outcome. *Experimental Design:* Patients with *BRAF* V600 mutation–positive melanoma were enrolled in each study after central confirmation of *... περισσότερα »

Prostate Cancer Stem Cell Therapy

Alexandros G. SfakianakisστοΟγκολογίαΠριν από 2 ώρες
*Purpose:* Clinical evidence suggests increased cancer stem cells (CSCs) in a tumor mass may contribute to the failure of conventional therapies because CSCs seem to be more resistant than differentiated tumor cells. Thus, unveiling the mechanism regulating CSCs and candidate target molecules will provide new strategy to cure the patients. *Experimental design:* The stem-like cell properties were determined by a prostasphere assay and dye exclusion assay. To find critical stem cell marker and reveal regulation mechanism, basic biochemical and molecular biologic methods, such as qu...περισσότερα »

Large Genomic Rearrangement in Pancreatic Ductal Tumors

Many somatic mutations have been detected in pancreatic ductal adenocarcinoma (PDAC), leading to the identification of some key drivers of disease progression, but the involvement of large genomic rearrangements has often been overlooked. In this study, we performed mate pair sequencing (MPseq) on genomic DNA from 24 PDAC tumors, including 15 laser-captured microdissected PDAC and […]

Inflammation in gastric cancer: Interplay of the COX-2/PGE2 and TLR/MyD88 pathways

Abstract Cyclooxygenase 2 (COX-2) and its downstream product prostaglandin E2 (PGE2) play a key role in generation of inflammatory microenvironment in tumor tissues. Gastric cancer is closely associated with Helicobacter pylori infection, which stimulates innate immune responses through Toll-like receptors (TLRs), inducing COX-2/PGE2 pathway through NF-κB activation. A pathway analysis of human gastric cancer shows […]

Applied Sciences, Vol. 6, Pages 39: Frequency-Dependent Amplitude Panning for the Stereophonic Image Enhancement of Audio Recorded Using Two Closely Spaced Microphones

Alexandros G. SfakianakisστοΝοσολογίαΠριν από 2 ώρες
In this paper, we propose a new frequency-dependent amplitude panning method for stereophonic image enhancement applied to a sound source recorded using two closely spaced omni-directional microphones. The ability to detect the direction of such a sound source is limited due to weak spatial information, such as the inter-channel time difference (ICTD) and inter-channel level difference (ICLD). Moreover, when sound sources are recorded in a convolutive or a real room environment, the detection of sources is affected by reverberation effects. Thus, the proposed method first tries to...περισσότερα »
