BRAF mutation and its inhibitors in sarcoma treatment
BRAF structures and their activation mechanisms. Abstract The mitogen‐activated protein kinase (MAPK ) signaling pathway plays a significant role in mediating cellular physiological activities, such as proliferation, differentiation, apoptosis, and senescence. This signaling pathway is composed of several major proto‐oncogenes of RAS/RAF/MEK/ERK , among which the BRAF proto‐oncogene, as one of the three members of the RAF family, has a higher mutation rate than ARAF and CRAF and has attracted...

MRI‐based radiomics nomogram to predict synchronous liver metastasis in primary rectal cancer patients
In the present study, we aimed to evaluate the predictive performance of radiomics nomogram for the diagnosis of synchronous liver metastasis in rectal cancer patients. Abstract At the time of diagnosis, approximately 15%‐20% of patients with rectal cancer (RC) presented synchronous liver metastasis (SLM), which is the most common cause of death in patients with RC. Therefore, preoperative, noninvasive, and accurate prediction of SLM is crucial for personalized treatment strategies. Recently, radiomics...

A measure of case complexity for streamlining workflow in multidisciplinary tumor boards: Mixed methods development and early validation of the MeDiC tool
There is increasing emphasis in cancer care globally for care to be reviewed and managed by multidisciplinary teams (ie, in tumor boards). Evidence and recommendations suggest that the complexity of each patient case needs to be considered as care is planned, however no tool currently exists for cancer teams to do so. We report the development and early validation of such a tool. We used a mixed‐methods approach involving psychometric evaluation and expert review to develop the Measure of case‐Discussion...

Noncoding RNA 886 alleviates tumor cellular immunological rejection in host C57BL/C mice
Nc886 broadly decreases the expression of some human leukocyte antigen molecules and antigen transporters, establishing the interesting role in regulating tumor cell antigens. Nc886 alleviates the inflammatory response of acute rejection caused by human‐derived prostate cancer cellular xenografts in immunocompetent mouse models. Nc886 reduces the polarization of macrophages through TGF‐β1 inhibition. Abstract Non‐coding RNA 886 (nc886/VTRNA2‐1) is a Pol III transcript and an atypical imprinted...

Genetic ablation of mast cells redefines the role of mast cells in skin wound healing and bleomycin-induced fibrosis
Conclusive evidence for the impact of mast cells (MCs) in skin repair is still lacking. Studies in mice examining the role of MC function in the physiology and pathology of skin regenerative processes have obtained contradictory results. To clarify the specific role of MCs in regenerative conditions, here we used a recently developed genetic mouse model that allows conditional MC ablation to examine MC-specific functions in skin. This mouse model is based on the cell type-specific expression of Cre...

Refined diagnostic criteria and classification of mast cell leukemia (MCL) and myelomastocytic leukemia (MML): a consensus proposal
Mast cell leukemia (MCL), the leukemic manifestation of systemic mastocytosis (SM), is characterized by leukemic expansion of immature mast cells (MCs) in the bone marrow (BM) and other internal organs; and a poor prognosis. In a subset of patients, circulating MCs are detectable. A major differential diagnosis to MCL is myelomastocytic leukemia (MML). Although criteria for both MCL and MML have been published, several questions remain concerning terminologies and subvariants. To discuss open issues,...

Mast cells are activated by Staphylococcus aureus in vitro but do not influence the outcome of intraperitoneal S. aureus infection in vivo
Staphylococcus aureus is a major pathogen that can cause a broad spectrum of serious infections including skin infections, pneumonia and sepsis. Peritoneal mast cells have been implicated in the host response towards various bacterial insults and to provide mechanistic insight into the role of mast cells in intraperitoneal bacterial infection we here studied the global effects of S. aureus on mast cell gene expression. After co-culture of peritoneal mast cells with live S. aureus we found by gene...

Proposed diagnostic algorithm for patients with suspected mastocytosis: a proposal of the European Competence Network on Mastocytosis
Mastocytosis is an emerging differential diagnosis in patients with more or less specific mediator-related symptoms. In some of these patients, typical skin lesions are found and the diagnosis of mastocytosis can be established. In other cases, however, skin lesions are absent, which represents a diagnostic challenge. In the light of this unmet need, we developed a diagnostic algorithm for patients with suspected mastocytosis. In adult patients with typical lesions of mastocytosis in the skin, a...

A New Numerical Approximation Method for Two-Dimensional Wave Equation with Neumann Damped Boundary
In this paper, a fully discretized finite difference scheme is derived for two-dimensional wave equation with damped Neumann boundary condition. By discrete energy method, the proposed difference scheme is proven to be of second-order convergence and of unconditional stability with respect to both initial conditions and right-hand term in a proper discretized norm. The theoretical result is verified by a numerical experiment.

Two Thalamic Regions Screened Using Laser Capture Microdissection with Whole Human Genome Microarray in Schizophrenia Postmortem Samples
We used whole human genome microarray screening of highly enriched neuronal populations from two thalamic regions in postmortem samples from subjects with schizophrenia and controls to identify brain region-specific gene expression changes and possible transcriptional targets. The thalamic anterior nucleus is reciprocally connected to anterior cingulate, a schizophrenia-affected cortical region, and is also thought to be schizophrenia affected; the other thalamic region is not. Ovarian Solid Pseudopapillary Tumor Resembling Benign Hemorrhagic Cyst on Rapid Frozen Section
Solid pseudopapillary tumors are rare, with the majority of described cases originating in the pancreas. To date, there are only 10 documented reports of primary ovarian solid pseudopapillary tumors. Here, we describe the case of a 24-year-old woman who presented with worsening pelvic pain and dysmenorrhea. Workup demonstrated a right ovarian solid mass on ultrasound and an elevated serum LDH, which raised concerns for dysgerminoma due to her relatively young age. Therefore, she was taken to the...

Here, we adopted a 3D high-frequency ultrasound imaging approach specifically for the investigation of CBI in human knee samples. First, an inhomogeneous intensity clustering energy is defined based on global distribution characteristics of the image intensities, and membership functions of the clusters described by the level set...

Long Noncoding RNA NEAT1 Regulates TGF-β2-Induced Epithelial-Mesenchymal Transition of Lens Epithelial Cells through the miR-34a/Snail1 and miR-204/Zeb1 Pathways
The aim of this study was to explore whether the long noncoding RNA nuclear paraspeckle assembly transcript 1 (NEAT1)/miR-34a/Snail1 and NEAT1/miR-204/Zeb1 pathways are involved in epithelial-mesenchymal transition (EMT) of lens epithelial cells (LECs). Primary human LECs (HLECs) were separated and cultured. Our results identified that TGF-β2 induces NEAT1 overexpression in a dose-dependent manner and a time-dependent manner. Additionally, TGF-β2 induced downregulation of E-cadherin and upregulation...
