Effect of cigarette smoke extract on lung epithelial cells and intervention on glutathione

Abstract: Objective To observe the morphological change and proliferation inhibition of A549 cells stimulated in vitro with different concentrations of cigarette smoke extract(CSE)and to explore the effect of smoking on lung epithelial cells and its reverse effect on glutathione by detecting the superoxide dismutase(SOD)activity and malondialdehyde(MDA)level.Methods In this study,the A549 cells were cultured by the nutrient solution of cigarette smoke extract with the concentration of 0%,2.5%,5.0%,7.5%,10.0% and 12.5%.Cell viability was measured by MTT method and cell proliferation morphology was determined by phase contrast microscope.Based on the above results,the experiment was divided into blank control group,2.5% CSE group,5.0% CSE group,7.5%CSE group and 7.5% CSE+GSH group(GSH 1 mmol/L).The SOD activity and MDA level were detected by colorimetric assay.Results 1After stimulation by CSE,the A549 cell showed decreased growth density,increased number of floating cells,atrophic adherent cells and inhibited proliferation.2After stimulation by CSE,the A549 cells were induced to dose-dependently secret MDA,which can lower the activity of SOD.3The MDA level in 7.5%CSE + GSH group were significantly lower than that in 7.5% CSE group,however,the SOD activity was significantly higher in 7.5% CSE + GSH group than in 7.5% CSE group.Conclusion 1The cigarette smoke extract can inhibit the growth of alveolar epithelial cells in a dose and time dependent manner.2The cigarette smoke extract can result in an oxidative damage on lung epithelial cells.3The glutathione could make a reverse effect of the oxidative damage caused by smoking.
