Abstract: AIM: To investigate the inhibitory effect of icariin on root resorption induced by rapid palatal expansion in rats. METHODS:24 male Wistar rats were divided randomly into 3 groups( n = 8) as group A( negative control),B( positive control) and C( experimental). In group B and C,an initial force of 50 g was applied to the area between the right and left upper first molars of the rats for 21 d for palatal expansion. The rats in group C were given intragastric administration of icariin at 2. 5 mg / kg / d while the rats in group A and B were given the same dose of placebo. 21 days after treatment the rats were sacrificed and the centered buccal-lingual root and peridentium tissue slices were generated from the upper first molars of the rats and were examined with HE and tararate- resistant acid phosphatase staining. Osteoprotegerin( OPG) and receptor activator of nuclear factor kappa- B ligand( RANKL) expression was determined by immunohistochemistry. RESULTS: Compared with group A and B,the number of osteoclass in group C decreased( P < 0. 05),the OPG / RANKL expression increased( P < 0. 05). CONCLUSION: Icariin may inhibit rapid palatal expansion- induced root resorption through increasing OPG / RANKL expression and decreasing osteoclast formation.
